摘 要:空间稳定免疫脂质体是将抗体或配体连接到修饰脂质体的聚乙二醇或其衍生物上,从而获得长循环和主动靶向的双重作用,在提高药效的同时还可减轻药物的不良反应。空间稳定免疫脂质体作为给药载体,对于毒性较大且需长期使用的抗肿瘤药、基因治疗药物和不易透过血脑屏障的药物表现出优异的输送性能,在临床治疗中有着广阔的应用前景。本文概括介绍了免疫脂质体发展的3个阶段,对空间稳定免疫脂质体的作用特点、分类以及抗体或配体连接脂质体的方法进行总结,并对其作为药物载体的应用情况进行归纳。Sterically stabilized immunoliposomes are developed from stabilized liposomes with polyethylene glycol (PEG) or its derivatives by coupling antibodies or ligands. This product has the dual function of long circulation and active targeting, so it can improve drug efficacy and reduce side effects. For delivery of antitumor drugs with bigger toxicity and for long-term use, gene therapy drugs and drugs which are hardly to pass through blood-brain barrier, sterically stabilized immunoliposomes show an excellent performance and have a broad prospect of application in clinical treatment. This review describes three stages of the development of immunoliposomes briefly, introduces the functional characteristics, classification and the methods of antibodies or ligands connecting to liposomes and summarizes the application of sterically stabilized immunoliposomes.
