摘 要:目的了解不同来源肠致病性大肠埃希菌(EPEC)分离株的血清型。方法采用特异性PCR方法检测大肠埃希菌不同O抗原编码基因,并以全套O抗血清复核确定O血清群;以PCR扩增和测序H抗原编码基因,经序列比对后确定H型。结果 331株EPEC分离株中,共检测到81种O血清型,其中优势O血清群为O51,占6.0%(20/331),O145占4.5%(15/331);共检测到25种H型,其中优势H型为H11(8.2%)、H2(7.9%)和H19(7.9%)。最常见的血清型为O51∶H7和O26∶H11。来自腹泻患者、健康从业人员、动物和生肉类的EPEC分离株中,分别检测到63种、22种、25种和17种O血清群。腹泻患者、动物和生肉类中的优势血清群/型分别为O51(8.7%)、O145(15.7%)和O76∶H7(21.2%),而健康人员的分离株无明显优势血清群。结论我国EPEC菌株的血清型呈现极大的多样性,无明显的优势血清群(型),传统上依靠血清群(型)对EPEC进行判断的方法存在局限性。Objective To identify the serotypes of enteropathogenic Escherichia coli( EPEC) strains isolated from different sources in China. Methods Methods Different O antigen encoding genes of E. coli were detected by specific PCR,and O serogroups were confirmed with a complete set of O antiserum. H antigen encoding genes were amplified and sequenced by PCR,and then confirmed by comparing sequences. Results In this study,81 different O serogroups and 25 H types were detected in the331 EPEC strains. The prominent O serogroups were O51( 20 /331,6. 0%) and O145( 15 /331,4. 5%). The prominent H types were H11( 8. 2%),H2( 7. 9%) and H19( 7. 9%). The most common serotypes were O51∶H7 and O26∶H11. The strains from diarrheal patients,healthy carriers,animals and raw meats presented 63,22,25 and 17 O serogroups,respectively. The most common O serogroups among strains from diarrheal patients,animals and raw meats were O51( 8. 7%),O145( 15. 7%) and O76∶H7( 21. 2%),respectively. But no significant common O serogroup was found in healthy carrier isolates. Conclusion These findings suggest that in China,the serotypes of EPEC strains are highly heterogeneous with no significant prominent O serotypes / serogroup being detected. There is great limitation for the traditional method defining EPEC by serology.
