摘 要:目的观察回医烙灸对兔膝骨性关节炎软骨细胞Bcl-2(B-cell lymphomas,B淋巴细胞-2蛋白)和Bax(Bcl-2 Associated X protein,B淋巴细胞-2的相关X蛋白)的表达的影响。方法将新西兰兔30只,随机分为正常对照组、模型对照组和回医烙灸组,每组各10只。采用Hulth造模方法制备兔膝骨性关节炎模型。回医烙灸组在造模4周后,在膝关节局部行回医烙灸疗法,1次/d,20 min/次,治疗4周。正常对照组和骨关节炎模型组不予干预。3组动物8周后处死,进行标本的大体形态学和光镜检测,Bcl-2、Bax蛋白的表达采用免疫组化方法进行检测。结果回医烙灸组关节软骨的组织学评分、软骨细胞Bax的表达灰度低于模型组(P<0.05),软骨细胞Bcl-2的表达灰度高于模型组(P<0.05)。结论回医烙灸疗法可以调节兔膝骨性关节炎软骨细胞Bcl-2和Bax的表达平衡,从而延缓膝骨性关节炎软骨的退变。Objective To observe the effect of moxibustion in Huiyi on articular chondrocyte apoptosis Bcl-2 and Bax of osteoarthritis rabbits. Methods A total of 30 adult New Zealand rabbits were randomly divided into normal group, animal model group and moxibustion group. Each group was 10 rabbits. The experimental knee osteoarthritis model was constructed by Hulth. The animals of moxibustion group were treated with moxibustion from 4 weeks,1/d, 20 min/times, 4 weeks of treatment.Normal control group and osteoarthritis model group were not treated,3 groups of animals were killed 8 weeks later. The changes of chondrocyte were observed by HE dyeing.The Bcl-2 and Bax was detected by immunohistochemical. Results The Bcl-2 change in moxibustion group was better than one in the animal model group(P 0.05). The expression of Bax was higher than in moxibustion group compared with animal model group(P 0.05). Conclusion Moxibustion in Huiyi can control the expression of Bcl-2 and Bax in the experimental knee osteoarthritis model. It maybe delay and inhibit the degeneration of chondrocyte.
