摘 要:目的观察益气活血方对破裂型腰椎间盘突出大鼠椎间盘组织P38信号转导通路信号通路的影响,初步探讨其改善的分子机制。方法建立腰椎间盘突出重吸收大鼠模型,大鼠随机分为模型组、益气活血方组、益气活血方+P38抑制剂组和P38抑制剂组,另设假手术组(n=10),假手术组及模型组灌胃纯净水,其他各组大鼠分别灌胃相应的药物,干预3周,3周后处死大鼠,测定血清中TXB2和PGF1α的含量和椎间盘组织中P-p38MAPK及MMP-3、MMP-7蛋白表达。结果模型大鼠经益气活血方治疗后,其血清TXB2含量和TXB2/PGF1α比值均明显下降,PGF1α含量升高,均呈良性变化;与模型组相比较,P<0.01。与抑制剂组比较,益气活血方+抑制剂组P-p38MAPK和MMP-3、MMP-7蛋白表达均增高(P<0.05或P<0.01)。结论益气活血方治疗后病变程度有所改善,其作用机制可能激活p38MAPK信号通路而提高椎间盘组织MMP-3、MMP-7蛋白表达有关。Objective To observe the influences of Yiqi Huoxue decoction on intervertebral disc and P38 signal pathway in rat model of ruptured lumbar disc,and to explore its preliminary molecular mechanism. Methods To establish a rat model of lunbar intervertebral disc and then all rats were divided into model group, Yiqi Huoxue decoction group, Yiqi Huoxue + blocker group and blocker group, and a sham operation group(n =10). The sham operation group and model group were intragastrically given purified water and other groups were given corresponding drugs for three weeks. The rats were killed after three weeks, the content of TXB2 and PGF1α in serum were detected, the expressions of P-p38 MAPK and MMP-3,MMP-7 in intervertebral disc were detected. Results After treatment, the serum levels of TXB2 and TXB2/PGF1 in the model rats were significantly decreased, and the content of PGF1 was increased, which showed a positive change. Compared with model group, the P 0.01, significant difference in the result. Content of P-p38 MAPK and MMP-3,MMP-7 phosphorylation level increased in Yiqi Huoxue decoction+ blocker group compared with blocker group(P 0.05 or P 0.01). Conclusion By Yiqi Huoxue decoction treatment, the lesions improved ratings have dropped which may be related to increasing phosphorylation levels of MMP-3 and MMP-7 by activation of P38 signal transduction pathway.
