摘  要:目的探讨苦瓜总皂苷对2型糖尿病大鼠心功能及心肌组织中脂联素受体1(Adipo R1)表达的影响。方法 60只雄性SD大鼠随机分为正常组,模型组以及苦瓜总皂苷治疗组,模型建立成功后,苦瓜总皂苷治疗组给予苦瓜总皂苷(20 mg/kg.d)灌胃治疗,对照组和模型组给予等量生理盐水灌胃,分别于治疗4、8周后处死大鼠各半,测定血糖、胰岛素、血脂以及心功能,HE染色检测心脏病理学,RT-PCR以及Western Blot检测心脏组织中Adipo R1 m RNA和蛋白的表达。结果造模成功后,糖尿病组大鼠血糖、血脂(TG、TC和FFA)以及心功能指标(CK、CK-MB、LDH和HBDH)明显升高(P<0.05),胰岛素含量明显降低(P<0.05),治疗4周后,除胰岛素明显升高外(P<0.05),各生化指标均明显下降(P<0.05),治疗8周后进一步改善;HE染色可见,糖尿病大鼠心脏有轻微的损伤,苦瓜总皂苷对其有一定的调节作用;造模成功后模型组心肌Adipo R1 m RNA水平和蛋白表达相较于正常组明显升高(P<0.05),苦瓜总皂苷治疗后其Adipo R1表达显著降低(P<0.05)。结论苦瓜总皂苷可通过降低2型糖尿病大鼠心脏组织中Adipo R1的表达,调节血脂,改善心脏功能,最终保护心肌细胞。Objective To invested the expression of total saponins of bitter gourd to cardiac function and adiponectin receptor 1(Adipo R1) of myocardial tissue in type 2 diabetic rats. Methods The 60 male SD rats were randomly divided into the normal group, model group and total saponins of bitter gourd treatment group, after the success built model, the treatment group given total saponins of bitter gourd 20mg/kg/d treated, respectively, after treatment of 4 weeks and 8 weeks half of rat were sacrificed, to detected of the blood sugar, insulin, blood lipid and cardiac function, HE staining detected the cardiac pathology, RT-PCR and Western Blot detected the Adipo R1 m RNA and protein expression. Results After the success of build the model, all group rats diabetes blood sugar, blood lipid and heart function were significantly higher(P 0.05), insulin levels significantly decreased(P0.05), after 4 weeks treated, except insulin increased significantly(P0.05), the others biochemical indicators were significantly decreased(P 0.05), and further improve after 8 weeks treatment; HE staining, diabetic rats cardiac damage slightly; after model built, Adipo R1 m RNA level and protein expression significantly increased compared with normal group(P〈0.05), Adipo R1 expression decreased significantly after treatment(P〈0.05). Conclusion Total saponins of bitter gourd can reduced the Adipo R1 expression in heart tissue of rats with type 2 diabetes, improve blood lipid and heart function, eventually protected myocardial cells.
