摘 要:目的:考察红景天苷对实验性高血压大鼠血管功能的改善作用。方法:胸主动脉缩窄法(TAC)制备高血压模型,观察长期连续给予红景天苷对大鼠血压、胸主动脉血管反应性、COX1及COX2表达水平的影响,并观察血管病理改变。结果:红景天苷干预治疗12周降低高血压大鼠血压值及死亡率,改善高血压大鼠胸主动脉对PE的反应性增强及对Ach内皮依赖性的血管舒张减弱;上调高血压大鼠血管COX1表达水平,抑制COX2表达,并能明显减少高血压引起的胸主动脉中膜厚度,减轻血管内皮损伤。结论:长期给予红景天苷治疗可明显改善高血压大鼠血管功能异常,有效降低血压,并可能通过抑制血管重构发挥心血管保护作用。Objective: To investigate the effects of long-term administration of 10mg/kg of Salidroside on vascular function in experimental hypertension rats. Methods: Male Sprague-Dawley (SD) rats were subjected to thoracic aorta coarctation (TAC) to induce hypertension. One day later, rats in Salidroside groups received an oral administration of 10 mg/kg of Salidroside per day for 12 weeks. The effects of Salidroside were valued by monitoring the blood pressure, determining the vascular functions of thoracic aorta, the expression of COXland COχ2 in thoracic aorta and the histopathological changes of aorta. Results: Compared with model rats, Salidroside treatment significantly lowered the rat mortality and the blood pressure, and ameliorated the vascular function of thoracic aorta as evidenced by inhibiting vascular contraction in response to PE and improving the vasodilation in response to Ach. While, the inhibitory effect of Salidroside on vasoconstriction was significantly reduced by pretreatment of indomethacin. Salidroside administration also increased COX1 expression, but inhibited the expression of COχ2 in aorta, which was reduced and increased in model group, respectively. Furthermore, the increased aortic intima-media thickness and vascular endothelial injury induced by hypertension were either reduced by long-term administration of Salidroside. Conclusion: Long-term administration of Salidroside can reduce blood pressure and exert cardiovascular protective effects possible through inhibiting the vascular remodeling and improving vascular functions
