复方甘西鼠尾注射液对大鼠心肌缺血再灌注损伤的保护作用

摘  要:目的与复方丹参注射液(丹参+三七)比较,探讨复方甘西鼠尾(甘西鼠尾+三七)对大鼠心肌缺血再灌注损伤的保护作用。方法采用在体和离体缺血再灌注模型,检测血清和灌流液中丙二醛(MDA)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH.Px)含量及在体心肌的梗死面积。结果复方甘西鼠尾注射液能显著降低在体和离体缺血再灌注后血清中MDA、CK、LDH水平,升高SOD和GSH—Px,缩小心肌的梗死面积,增加心脏冠脉流量,作用相似于复方丹参。结论复方甘西鼠尾注射液有较好的抗心肌缺血再灌注损伤作用,其机制可能与抗氧自由基损伤有关。Objective To find out the cardio protection induced by the compounded injections made from Salvia przewalskii Maxim (SP) + Panax notoginseng (Burk.) F.H.Chen (PN) during ischemia/reperfusion in rats, compared with injection made from Salvia miltiorrhiza Bunge (SM) + PN, and to find out whether SP can be used as SM against cardio ischemia/reperfusion injury or not. Methods Two ischemia/reperfusion models were employed. In one model, we ligated rat's artery of left anterior descending (LAD) for 30 min, then let it loose for 60 min, we examined maleic dialdehyde (MDA), lactate dehydrogenase (LDH), creatine kinase (CK), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) in rats' serum after reperfusion, and the infarct zone sizes were checked. In another model, in vitro, we perfused every rat's heart at low rate of flow with Kreb's solution for 15 min at first, then followed normal perfusion for 30 min also examined MDA, LDH, CK, SOD, and GSH- Px in the flow of effluent after reperfusion. Results The injection of SP+PN is able to decrease MDA, CK, LDH, and increase SOD, GSH-Px, in vivo and in vitro, the infarct zone sizes were significantly alleviated, and increase coronary flow, not less powerful than that of SM+PN. Conclusion The compounded injection from SP+PN employed in cardio protection against ischemia/reperfusion injury, is not less powerful than the injection from SM+PN, which suggests SP can be a likely replacer for SM to protect myocardium from ischemia/reperfusion injury. Its mechanisms may be associated with deactivating oxygen free radicles.
