青鹏膏剂穴位贴敷对大鼠肥大细胞数量和脱颗粒率的影响

摘　　要：目的:观察青鹏膏剂大椎穴贴敷对大鼠穴区组织肥大细胞(MC)数量和脱颗粒率的影响。方法:雄性Wistar大鼠30只随机分为青鹏膏剂穴位贴敷组、电针组和空白对照组,每组10只。青鹏膏剂组大鼠大椎穴穴区局部剃毛后涂抹青鹏膏剂(约25mg,2次/天,每次20min),电针组大鼠大椎穴给予电针刺激。采用甲苯胺蓝染色观察MC的形态、数量和脱颗粒率。结果:青鹏膏剂组和电针组大鼠穴区组织内MC细胞数量增多、体积增大,细胞膜破裂,细胞轮廓不规则,MC脱颗粒现象明显,且MC多沿神经或血管分布。青鹏膏剂组和电针组大鼠穴区组织MC数量比较差异无统计意义(P>0.05),但均明显高于空白对照组(P<0.05)。三组大鼠穴区组织脱颗粒的MC个数和MC脱颗粒率由高到低依次为青鹏膏剂组、电针组、对照组,两两比较差异具有统计学意义(P<0.05)。结论:青鹏膏剂穴位贴敷可激活MC,出现数量增多及募集和脱颗粒现象。Objective： To observe the effects of Qingpeng Unguenturn Dazhui point sticker on number and degranulation rate of mastocyte in rats. Methods： 30 male Wistar rats were randomly divided into blank control group, electroacupuncture group and Qingpeng Unguentum group with 10 rats in each group. The Dazhui point of rats in Qingpeng Unguentum group were stuck with 25mg Qingpeng Unguentum for 2 times a day; The Dazhui point of rats in electroacupuncture group were stimulated with electroacupuncture. Toluidine blue staining was used to observe the morphology, number and degranulation of mastocyte. Result： In Dazhui point area, the number and volume of mastocytes of rats in electmacupuncture group and Qingpeng Unguenturn group increased; The cell membrane ruptured and mastocytes showed obviously degranulation; Moreover, the mastocytes distributed along nerves or blood vessels. The number ofmastocytes in Dazhui point area of rats in electroacupuncture group and Qingpeng Unguentum group were obviously more than conlrol group （P〈0.05）. The quantity and ratio of degranulated mastocytes from high to low were Qingpeng Unguentum group, electroacupuncture group and control group, there had statistically significant differences between every 2 groups （P〈0.05）. Conclusions： Qingpeng Unguentum acupoint sticker can activate mastocyte, which show number increasing and degramtlation.
