摘  要:目的观察坤舒康颗粒对大鼠棉球肉芽肿的抑制作用,初步探讨其作用机制。方法将SD大鼠随机分为假手术组、模型组、阳性药地塞米松组(阳性组)及坤舒康颗粒高剂量组、中剂量组、低剂量组,每组10只。除假手术组外,其余大鼠麻醉后,于左前肢腋下植入灭菌棉球20 mg。坤舒康各剂量组大鼠分别灌胃不同剂量坤舒康混悬液(20、10、5 g/kg体重),阳性药组大鼠灌胃地塞米松混悬液(0.08 g/kg体重),连续给药14 d。末次给药后麻醉,取出棉球,干燥称重;取胸腺、脾脏,称重,计算胸腺、脾脏指数;股动脉取血,分别检测血浆6-酮-前列腺素F_(1α)(6-keto-PGF_(1α))、血栓烷B_2(TXB_2)及血清白介素-1β(IL-1β)、一氧化氮(NO)水平。结果与假手术组比较,模型组大鼠的胸腺及脾脏指数明显降低(P<0.01),血浆6-keto-PGF_(1α)、TXB_2及血清IL-1β、NO水平均明显升高(P<0.01)。与模型组比较,坤舒康颗粒高剂量组棉球肉芽肿重量明显降低(P<0.05),胸腺、脾脏指数则无明显变化(P>0.05),坤舒康颗粒高剂量、中剂量组大鼠血浆6-keto-PGF_(1α)、TXB_2及血清IL-1β、NO水平均明显降低(P<0.01或P<0.05)。结论坤舒康颗粒对慢性肉芽肿性炎症具有抑制作用,其抗炎作用机制与抑制炎症介质释放有关。Objective To study the inhibitive effect and possible mechanism of Kunshukang (KSK) Granules on cotton ball granuloma model. Methods Sixty SD rats were randomly divided in t9 6 groups: control group, mode! group, Dex- amethasone group, high dose KSK group, middle dose KSK group and low dose KSK group. All groups except control group rats were implanted 20 mg cotton ball in left armpit. The rats in every dose KSK groups were administrated KSK suspension (20, 10, 5 g/kg), and the rats in Dexamethasone group were administrated Dexamethasone suspension(0.08 g/kg) for 14 days. Then the cotton ball were removed, dried and weighed. All rats were fetched and weighed thymus and spleen, taken blood into analysis 6-keto-PGF, TXB2, IL-1β and NO. Results Compared with sham-operated group rats, the thymus and spleen index of model group rats were significantly reduced (P 〈 0.01), and the level of 6-keto- PGFI,TXB2, IL-1β and NO of model group rats were significantly raised (P 〈 0.01). Compared with model group rats, the cotton ball weight of high dose KSK group rats were significantly reduced (P 〈 0.05), and the level of 6-keto- PGF, TXB2, IL-1β and NO of high and middle dose KSK groups were significantly reduced (P 〈 0.01 or P 〈 0.05).Conclusion Kunshukang Granules can inhibit cotton ball granuloma by inhibiting the release of inflammatory mediators.
