摘  要:目的观察旋转刺激对大鼠的内毒素敏感性及脑干神经递质含量的影响,旨在为海上环境下的抗感染治疗提供依据。方法采用仿制Crampton旋转刺激器建立大鼠模拟运动病模型。将104只大鼠随机均分为4组:正常对照组、旋转刺激组、内毒素损伤组、旋转刺激+内毒素损伤组。其中旋转刺激组仅给予旋转刺激90 min,内毒素损伤组给予3 mg/kg内毒素脂多糖(lipopolysaccharide,LPS)腹腔内注射,旋转刺激+内毒素损伤组予内毒素腹腔注射后再接受旋转刺激。1生存率观察:每组20只,分别观察各组72 h内大鼠的一般状态及生存情况。2中枢神经递质测定:每组6只,检测腹腔注射后2 h时点的各组脑干组织乙酰胆碱(acetylcholine,Ach)和去甲肾上腺素(norepinephrine,NE)含量。结果172 h生存率比较,正常对照组和旋转刺激组均无大鼠死亡,内毒素损伤组的生存率为85%,明显高于旋转刺激+内毒素损伤组55%的生存率,组间比较差异具有统计学意义(P<0.05)。2脑干组织的神经递质含量比较,旋转刺激组的Ach含量较正常对照组明显升高,组间比较差异具有统计学意义(P<0.05),但内毒素损伤组和旋转刺激+内毒素损伤组的变化不明显。旋转刺激组、内毒素损伤组以及旋转刺激+内毒素损伤组的NE含量均较正常对照组明显下降,组间比较差异具有统计学意义(P<0.05)。结论旋转刺激可明显增加大鼠对内毒素损伤的敏感性,其机制可能与脑干胆碱能和NE能系统的功能紊乱有关。Objective To explore the effects of rotatory stimulation to the endotoxin damage on the sensitivities of rats induced by lipopolysaccharide(LPS),as well as the contents of neurotransmitters in brainstem tissues,so as to provide a basis for anti-infective therapy during deployment at sea.Methods The rat model of motion sickness was established by using the generic copies of Crampton rotatory simulator.A total of 104 rats were randomly divided into four groups:normal control group,rotatory stimulation group,endotoxin injury group and rotatory stimulation plus endotoxin injury group.Rats were received 90 minutes rotatory stimulation in rotatory stimulation group,and rats in endotoxin injury group were given intraperitoneal injection with a nonlethal dose of LPS(3 mg/kg),while rats in rotatory stimulation plus endotoxin injury group were received rotatory stimulation after LPS injection.Two tests were made in the study,one was survival rate test,and the other was central neurotransmitter test.In the survival rate test,the general status and survival rate of rats was observed within72 hours in each group(n=20).While in the central neurotransmitter test,contents of acetylcholine(Ach)and norepinephrine(NE)in brainstem tissue were detected respectively in the second hour with intraperitoneal injection in each group(n=6).Results Both normal control group and rotatory stimulation group had no rats died,the survival rate(55%)in rotatory stimulation plus endotoxin injury group was significantly lower than that(85%)in endotoxin injury group(P〈0.05).Compared with normal control group,the contents of Ach in the brainstem tissues increased significantly in rotatory stimulation group(P〈0.05),but no difference was showed in both endotoxin injury group and rotatory stimulation plus endotoxin injury group(P〉0.05).The contentss of NE decreased markedly in all the three groups as compared with that in normal control group(P〈0.05).Conclusion The rotatory stimulation may increse
