摘 要:以紫花苜蓿中苜1号为研究对象,采用固相萃取(SPE)纯化后紫外分光光度(UV)法对紫花苜蓿不同组织中的总皂甙含量进行比较研究。通过对总皂甙提取纯化工艺进行优化,以香草醛-高氯酸为显色剂使皂甙显色后,用分光光度法(λ=545nm)对紫花苜蓿根、茎、叶、种子及全株总皂甙含量进行测定。结果表明,最佳提取纯化工艺为:70%乙醇溶液进行超声辅助提取、提取液注入Sep-Pak C18后用水和30%甲醇淋洗纯化,最终4ml甲醇洗脱总皂甙。紫花苜蓿不同部位总皂甙含量分布不均匀:根1.98%~2.27%、茎0.87%~0.97%、叶1.15%~1.30%、种子0.88%~0.99%、全株1.08%~1.23%。Medicago sativa L. cv. Zhongmu No. 1 was used as the object of the research, saponins in different tissues of alfalfa were measured with UV speetrophotometry using solid phase extraction(SPE). The extraction--purification process was optimized, the total alfalfa saponins in root, stem, leaf, seed and whole herb were determined with spectrophotometry (λ=545 nm). The results showed that the optimal extraction and purification process was: ultrasonic assisted extraction with 70G ethanol, followed by SPE C18 Sep-Pak cleanup, the SPE cartridge washed with water and 30% methanol and saponins eluted with 4 ml of 100% methanol. The saponins content in Medicago sativa L. ‘Zhongmu No. 1'was 1.98-2.27%, 0.87-0.97%,1.15-1.30%,0.88-0.99%and 1.08-1.23%in root, stem, leaf, seed and whole herb, respectively.
