心肌缝隙连接蛋白43在运动预处理不同保护时相表达变化的比较研究

摘  要:目的:比较大鼠心肌缝隙连接蛋白43(Cx43)mRNA和蛋白在运动预处理(EP)早、晚期保护时相的表达变化。方法:SD大鼠32只随分为对照组(C组)、力竭运动组(EE组)、早期运动预处理+力竭运动组(EEP+EE组)和晚期运动预处理+力竭运动组(LEP+EE组)。在建立EP和EE动物模型基础上,用原位杂交和实时荧光定量PCR方法观察和检测心肌Cx43 mRNA表达,用免疫组织化学和Western免疫印迹方法观察和检测心肌Cx43蛋白表达。结果:与EE组相比,EEP+EE和LEP+EE组心肌Cx43 mRNA和蛋白水平显著升高。与EEP+EE组相比,LEP+EE组心肌Cx43原位杂交信号没有显著变化,Cx43 mRNA表达也没有显著性差异。与EEP+EE组相比,LEP+EE组心肌Cx43免疫阳性反应无明显变化,Cx43蛋白表达也没有显著性差异。结论:运动预处理早、晚期保护时相分别促进了心肌Cx43 mRNA和蛋白的表达,但在这两个保护时相之间,Cx43 mRNA和蛋白的表达没有明显差异,表明心肌Cx43在介导运动预处理早、晚期心肌保护效应中的表达变化规律和运动预处理早、晚期保护时相的变化具有同步性。Objective To compare the expression of myocardial connexin 43(Cx43)mRNA and its pro-tein during early and late cardioprotection stages of exercise preconditioning. Methods Thirty- twoSprague-Dawley rats were randomly divided into a control group,an exhaustive exercise(EE)group,anearly exercise preconditioning + exhaustive exercise(EEP+EE)group and a late exercise preconditioning+ exhaustive exercise(LEP + EE)group,each of 8. All groups were given intervention as their groupname indicated. Then in situ hybridization and real- time fluorescent quantitative PCR methods wereused to detect the changes of myocardial Cx43 mRNA and immunohistochemical method and Westernblotting were used to detect the changes of Cx43 protein. Results Compared with EE group,there wassignificant increase in Cx43 mRNA and its protein expression in group EEP+EE and LEP+EE. Com-pared with EEP+EE group,no significant changes was found in situ hybridization and Cx43 Immunoreac-tivity in LEP+EE group,neither did significant differences in the expression of Cx43 mRNA and its pro-tein. Conclusion EEP and LEP can significantly promote the expression of myocardial Cx43 mRNAand its protein respectively. However there is no significant changes of myocardial Cx43 mRNA andprotein expression between the 2 time phases. It demonstrates that the expression of Cx43 in the earlyand late stage of myocardial protective effect was consistent with the changes of the early and late phase of the protective effect of EP.
