摘  要:【目的】组织工程中种子细胞需依赖于细胞外基质的存在才能发挥功能,因此支架材料的选择具有重大意义。【方法】为获得具有类似人骨的渐变孔结构、力学性能良好的仿生骨材料采用mPEG与PDLLA共聚制备改性PDLLA,并与nHAP/TTCP复合,通过控制造孔剂的粒径和用量得到不同孔径、孔隙率的mPEG-PDLLA/nHAP/TTCP有机无机复合骨支架。FT-IR考察共聚物结构,液体置换法测定支架孔隙率和密度,接触角测定法考察材料亲水性,与成骨细胞联合培养考察支架生物相容性。【结果】mPEG与PDLLA通过酯键共聚;mPEG改性PDLLA增加聚乳酸亲水性;支架的孔隙率在70%~90%,与成骨细胞相容性良好。【结论】mPEG-PDLLA/nHAP/TTCP可作为组织工程支架提供成骨细胞黏附与增殖,【Objective】Seed cells are dependent on the extracellular matrix in tissue engineering to carry out their functions, so choosing scaffold materials is of great significance.【Methods】In order to obtain tapered porous structure similar to human bone and bionic bone materials good at mechanical properties, mPEG and PDLLA were copolymerized to prepare modified PDLLA, then combined with nHAP/TTCP and obtained organic and inorganic composite bone scaffold of mPEG-PDLLA/nHAP/TTCP in different porosity and aperture by controlling the pore size and amount. FT-IR was used to determine the copolymer structure, while porosity rate and density were determined by liquid displacement method. Contact angle measurement method was used to investigate the hydrophilicity of the materials. Combined culture of scaffold and osteoblasts was used to determine the biocompatibility.【Results】Ester bond copolymerized mPEG and PDLLA; mPEG-modified PDLLA increased the hydrophilicity of poly lactic acid; the porosity rates of the scaffolds were between 70%-90%, and the compatibility with osteogenic cell was good.【Conclusion】mPEG-PDLLA/nHAP/TTCP can be used as scaffolds for tissue engineering and provide the adhesion and proliferation for osteoblasts.
