参苏饮对炎症人支气管上皮细胞表达β-防御素2的作用研究

摘　　要：目的:观察参苏饮对炎症人支气管上皮细胞(16HBE)表达β-防御素2(h BD-2)的影响。方法:将40只SPF级SD雄性大鼠灌胃给药参苏饮及其拆方,心脏采血制备含药血清,传代培养人支气管上皮细胞(16HBE),利用1mg/L LPS刺激12h建立体外16HBE细胞炎症模型后分为5组,即空白对照组、模型组、参苏饮全方组、益气解表组和止咳化痰组。Western法检测给药3h、8h后各组胞浆蛋白h BD-2的表达情况。结果:与空白对照组比较,模型组h BD-2蛋白含量在给药3h显著升高(P<0.05),而在给药8h两组比较差异无统计学意义(P>0.05);与模型组比较,参苏饮全方组、益气解表组、止咳化痰组在给药3h、8h均能显著性上调h BD-2蛋白含量(P<0.05)。结论:参苏饮能够促进炎症人支气管上皮细胞表达h BD-2蛋白,提示参苏饮发挥益气固表、抗菌消炎的物质基础可能与h BD-2有关。Objective ： To investigate the effect of Shensu drink on the expression of human β - defensin - 2 （ hBD - 2） in inflammatory human bronchial epithelialcells （ 16HBE ）. Methods ： A total of 40 specific pathogen - free Sprague - Dawley rats were given Shensu drink and its separated prescriptions by garage. Blood was taken from the heart to prepare drug -containing serum. Human bronchial epithelial cells （16HBE）were subcultured, 1 mg/L lipopolysaccharide stimulation for 12 hours was used to establish the in vitro16HBE inflammation model ,and then the 16HBE cells were divided into blank control group ,model group, Shensu drinkwhole prescription group, qi -tonifying and exterior syndrome -relieving group ,and cough -relieving and phlegm -eliminating group. Western blot was used to measure the expression ofcytoplas- mic protein hBD -2 in each group at 3 and 8 hours after administration. Results：Compared with the blank control group, the model group had a significant increase in the content of hBD - 2 at 3 hours after administration （P 〈 0. 05 ）, while there was no significant difference between these two groups at 8 hours after administration （ P 〉 0. 05 ）. Compared with the model group, the Shensu drinkwhole prescription group, the qi -tonifying and exterior syndrome -relieving group, and the cough - relieving and phlegm - eliminating group had significantly upregulated expression of hBD - 2 at 3 and g hours after administration （P 〈 0. 05 ）. Conclusion ： Shensu drink can promote the expression of hBD - 2 in inflammatory human bronchial epithelialcells,which suggests that the material basis of the qi -tonifying, exterior syndromerelieving, anti - microbial, and anti - inflammatory effects of Shensu drink may be related to hBD - 2.
