摘 要:目的观察加味良附丸联合5-Fu对人胃癌细胞SGC-7901裸鼠移植瘤生长抑制作用及其对肿瘤组织中核转录因子-kappaBp65(NF-ΚBp65)表达的影响。方法建立人胃癌细胞SGC-7901移植瘤动物模型,随机分为模型组、5-FU组及加味良附丸大、中、小剂量组、加味良附中剂量+5-FU组(联合给药组)。给药10 d,计算肿瘤抑制率,免疫组化检测肿瘤组织中NF-ΚBp65的表达。结果联合给药组、5-FU组、加味良附丸大、中、小剂量组抑瘤率分别为54%、45%、31%、28%、17%,联合给药组与加味良附丸大、中、小剂量比,差异显著有统计学意义(P<0.05);模型组肿瘤组织中NF-ΚBp65表达明显高于各给药组,差异显著有统计学意义(P<0.05);联合给药组肿瘤组织中NF-ΚBp65表达低于加味良附丸大、中、小剂量组,差异明显有统计学意义(P<0.05)。结论加味良附丸联合5FU组抑瘤率高于单纯加味良附丸高、中、低剂量组及5-FU组,联合给药抑制NF-ΚBp65表达方面优于加味良附丸高剂量组及5-Fu组,提示中西药联合具有协同作用。Objective To observe the effects of modified liang-fu formula (MLFF) combined with 5-Fu which focus on NF-KBp65 level in tumor tissues of SGC-7901 tumor bearing nude mice and the tumor growth inhibition. Methods Establish the tumor-bearing nude mice models with human gastric cancer cell line SGC- 7901, which were divided randomly into six groups : blank group,5-Fu group, MLFF in high, medium, low dose groups, and combination group. The mice were treated for 10 days and then detecting NF-KBp65 level in tumor tissues of SGC-7901 tumor bearing nude mice and calculateing the tumor inhibition ratio. Results The tumor inhibition ratio of combination group was 54%, which had significant difference between blank group ( P〈0.05. The tumor inhibition ratio of combination group was higher than MLFF in high dose group remarkable. The contents of NF-KBp65 in combination group was lower than blank group, 5- Fu group and MLFF in high dose group respectively. Conclusion The tumor inhibition ratio of combination group is higher than 5-Fu group and MLFF in high dose group, meanwhile, the combination group is better than 5-Fu group and MLFF in high dose group on regulating expression of NF-KBp65 level in tumor tissues. Which means Modified Liang-Fu Formula Combined with 5-Fu has synergistic effects.
