摘 要:目的了解浙江省不同地区淡色库蚊钠离子通道(vssc)基因多态性分布特征。方法采集杭州、舟山、湖州、金华等市区淡色库蚊样本,PCR扩增vssc基因片段,扩增产物进行基因单链构象多态性(single strand conformation polymorphism,SSCP)分析。对不同的SSCP型进行测序,并进行氨基酸序列预测。结果四地市淡色库蚊现场品系以及敏感品系vssc基因片段经SSCP分析共发现7个SSCP型,分别属于3个不同的氨基酸序列型,即野生型、L1014S突变型以及L1014F突变型。结论浙江省淡色库蚊vssc基因存在较高的多态性,使其在面对菊酯类等相关杀虫剂选择压力时具有较高的耐受度,进而可能会增大相关杀虫抗性防制的难度。Objective To study genetic polymorphism of voltage sensitive sodium channel (vssc) gene of Culex pipiens pallens in Zhejiang Province. Methods Mosquitoes of Cx. pipiens pallens were captured in Hang- zhou, Zhoushan, H uzhou, J inhua, and vssc genes were amplified by PCR amplification, and the single strand conformation polymorphism (SSCP) of vssc gene was analyzed. The sequence analysis was performed for differ- ent SSCP, and amino acid sequence was predicted. Results Total 7 SSCP types of vssc genes of Cx. pipiens pallens captured in four cities were identified, which belonged to 3 amino acid sequence types as wild type, L1014S mutant type and L1014F mutant type. Conclusions The vssc genes of Culex pipiens pallens in Zhe- jiang Province are highly variable, which makes mosquitoes more resistant to insecticides.
