复荣通脉胶囊对糖尿病大鼠心肌III型胶原蛋白及纤连蛋白表达的影响

摘　　要：目的观察复荣通脉胶囊对糖尿病大鼠心肌纤连蛋白（1aminin，LN）及Ⅲ型胶原蛋白（collagen III,Col III）表达的影响。方法按随机数字表法将50只大鼠分为正常对照组，模型组，复荣通脉胶囊低、中、高剂量组，各10只。成模后1周，低、中、高剂量组分别灌胃复荣通脉胶囊混悬液0．7、1．4、2．8g／kg，正常对照组、模型组大鼠灌胃等体积生理盐水。灌胃1次，d，连续给药8周。末次给药后，检测大鼠血清TC、TG水平；采用免疫组化法观察各组大鼠心肌组织LN及Col Ⅲ表达。结果与模型组比较，复荣通脉胶囊中、高剂量组LN[（0．67±0．04）％、（0．65±0．09）％比（1．08±0．13）％]、ColⅢ[（0．67±0．15）％、（0．69±0．13）％比（1．17±0．12）％]表达明显降低（P〈0．05）。复荣通脉胶囊各剂量组大鼠血糖及血清TC、TG水平与糖尿病组比较差异无统计学意义（P〉0．05）。结论复荣通脉胶囊可抑制糖尿病大鼠心肌LN及Col Ⅲ表达。Objective To explore the the influence of Furong-Tongmai capsules on myocardial expression of LN and Collagen Ⅲ in diabetes mellitus （DM） rats. Methods A total of 60 rats were randomly divided into the control group, the model group, the low-, middle- high-dosage Furong-Tongmai capsules group （n=10）. The low-, middle-high-dosage Furong-Tongmai group was given 1.4, 0.7, 2.8 g/（kg body weight） Furong-Tongmai capsules. The other two groups were given the same dose of purified water. After 8 weeks treatment, the myocardial was taken to make pathology slice with SP immunohistochemistry staining. The expression of LN and Collagen III were detected. Results Compared with model group, the expression of LN （0.67% ± 0.04%, 0.65% ± 0.09% vs. 1.08% ± 0.13%） and Collagen Ⅲ （0.67%± 0.15%, 0.69% ±0.13% vs. 1.17%±0.12%） in the middle-high-dosage Furong-Tongmai groups significantly decreased （P〈0.05）. However, there were no statistical differences between the low-, middle- high-dosage Furong-Tongmai groups and the model group （P〉0.05）. Conclusions The Furong-Tongmai capsules could inhibit the expression of LN and Collagen Ⅲ in DM rats.
