摘 要:目的:观察电针双侧"足三里"穴对脾气虚模型大鼠下丘脑及小肠组织胆囊收缩素(CCK)及其受体(CCK-AR)表达的影响。方法:随机将40只SD大鼠分为4组,即正常组、脾气虚组、足三里组和非经非穴组,每组10只。除正常组外,其余3组均采用力竭游泳和不规则饮食复合法建立脾气虚证大鼠模型。依据造模评定标准造模成功后,足三里组给予电针大鼠双侧"足三里"穴干预处理,采用疏密波,1次/天,20 min/次,连续6天;治疗结束后,10%水合氯醛腹腔麻醉,取下丘脑及小肠组织,采用酶联免疫吸附法(ELISA)检测大鼠小肠组织CCK含量变化,荧光定量PCR法检测大鼠小肠及下丘脑组织CCK及CCK-AR mRNA表达水平。结果:脾气虚组和非经非穴组下丘脑及小肠组织中CCK、CCKAR mRNA表达水平明显高于正常组(P<0.05);足三里组下丘脑及小肠组织中CCK及CCK-AR mRNA表达水平相比于脾气虚组降低(P<0.05),非经非穴组未见显著性差异(P<0.05)。结论:针刺"足三里"穴可以调节脾气虚大鼠小肠及下丘脑组织CCK及CCK-AR的异常分泌,发挥其外周和中枢效应对脾气虚证起调控作用。Objective: To observe the effect of electroacupuncture at bilateral "Zusanli"on the expression of cholecystokinin( CCK) and its receptor( CCK- AR) in small intestine and hypothalamus of spleen Qi deficiency model rats. Methods: 40 SD rats were randomly divided into four groups,normal group,spleen Qi deficiency group,Zusanli group and non- acupoint group,with 10 in each group. Except the normal group,other three groups received exhaustive swimming and irregular diet to establish the models of spleen Qi deficiency syndrome. Zusanli group was given interventive treatment by acupuncturing at bilateral "Zusanli"after modeling based on the standard for modeling evaluation,using density wave,1 time / d,20 min / time,lasting six days; after treatment,with 10% chloral hydrate intraperitoneal anesthesia,take out the small intestine and hypothalamus.The changes of CCK content in small intestine tissue of rats were detected by enzyme linked immunosorbent assay( ELISA). Detect CCK and mRNA CCK- AR expression in rats' small intestine and hypothalamus by fluorescence quantitative PCR. Results: The CCK and CCK- AR mRNA expression levels in hypothalamus and small intestine tissues in spleen Qi deficiency group and non- acupoint group were significantly higher than those in normal group( P〈0. 05); CCK and CCK- AR mRNA expression levels in hypothalamus and small intestine tissue were significantly decreased in Zusanli group compared with those in the spleen Qi deficiency group( P〈0. 05). Conclusion: Acupuncture at "Zusanli"can regulate the abnormal secretion of CCK and CCK- AR in small intestine and hypothalamus of rats with spleen Qi deficiency,playing a regulative role of peripheral and central action in the function of spleen Qi deficiency syndrome.
