摘  要:目的建立测定小儿氨酚烷胺颗粒中对乙酰氨基酚和盐酸金刚烷胺含量的HPLC-ELSD法。方法色谱柱为Gemini Phenomenex C_(18)(250mm×4.6mm,5μm);流动相为1.0mL·L^(-1)的三氟乙酸溶液-甲醇(70∶30);流速为1.0mL·min^(-1);柱温为25℃;蒸发温度为40℃;雾化温度为40℃;氮气载气流速为1.8L·min^(-1)。结果对乙酰氨基酚和盐酸金刚烷胺质量浓度分别在2.017~10.084和1.006~5.032 mg·mL^(-1)范围内峰面积与进样量的对数呈良好的线性关系(r值分别为0.999 8和0.999 6);平均回收率分别为99.8%和98.6%,RSD值分别为0.42%和0.41%。结论该方法简便、快速、准确且重复性好。Objective To establish a method for the determination of paracetamol and amantadine hydrochloride in Pediatric Paracetamol and Amantadine Hydrochloride Granules by HPLC-ELSD.Methods The chromatographic column was Gemini Phenomenex C18(250mm×4.6mm,5μm);the mobile phase was 1.0mL·L^-1 trifluoro acetic acid solution-methanol(70∶30);the flow rate was 1.0mL·min^-1;the column temperature was 25 ℃;the evaporation temperature was 40 ℃;the atomization temperature was40 ℃;flow rate of nitrogen carrier the gas was 1.8L·min^-1.Results The logarithm of the contents of paracetamol had good linear relationship with the logarithm of their peak area in the range of 2.017-10.084mg·mL^-1(r=0.999 8),and the logarithm of the contents of amantadine hydrochloride had good linear relationship with the logarithm of their peak area in the range of 1.006-5.032mg·mL^-1(r=0.999 6);the average recoveries were 99.8% and 98.6%(RSD=0.42% and 0.41%).Conclusion The method is proved to be simple,rapid,accurate and has excellent precision.
