独活寄生汤对兔膝骨性关节炎关节液中白介素-1β、肿瘤坏死因子-α和Ⅱ型胶原的影响

摘　　要：目的观察独活寄生汤对兔膝骨性关节炎关节液中白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和Ⅱ型胶原(CollagenⅡ)表达水平的影响,探讨其治疗骨性关节炎的机制。方法将40只实验用新西兰大白兔分五组,即空白对照组、模型组、独活寄生汤低剂量组、高剂量组和玻璃酸钠组,每组各8只,采用改良伸直位石膏固定兔膝关节造模,造模后立即干预,独活寄生汤组浓缩液给药,每日1次;对照组关节内注射玻璃酸钠注射液,0.5m L/关节,1周1次,造模8周,采用双抗体夹心酶免疫吸附法(ELISA)和免疫组化法,分别测定兔膝关节液中的IL-1β、TNF-α表达水平以及Collagen II的表达水平。结果兔膝骨性关节炎造模后,兔的Mankin评分明显升高(P<0.01),独活寄生汤和玻璃酸钠可降低Mankin评分(P<0.01),且两者的比较差异无显著意义(P>0.05)。结论独活寄生汤可以通过降低关节液中IL-1β、TNF-α的水平以及保护Collagen II以对兔膝骨性关节炎起到防治作用;Objective To observe the effects of Duhuo Jisheng decoction on the expression of IL-1β,TNF and Collagen II of synovial fluid in rabbits with osteoarthritis,and discuss its mechanism of osteoarthritis. Methods 40 New Zealand rabbits were randomly divided into 5 groups： blank control group,model group,low dose Duhuo Jisheng decoction group,high dose Duhuo Jisheng decoction group and Sodium hyaluronate group,eight in each group. OA animal models were made by fixing right posterior knee joint in stretching state with plaster cast,intervention was used at the beginning of model preparation. Duhuo Jisheng decoction groups were given medicine by concentrated liquid,once per day,control group was treated with joint cavity injection of Sodium hyaluronate（ 20 mg / m L,0. 5m L per inject,once per week）. At the 8th week after model preparation,concentrations of IL-1β and TNF-a in joint synovial fluid were detected by ELISA assay and Collagen II was detected by immunohistochemistry method. Results Mankin scores of rabbits increased after knee osteoarthritis（ P〈0. 01）,and the scores of Duhuo Jisheng decoction group and Sodium hyaluronate group were lower than that of model group（ P〈0. 01）,the scores in Duhuo Jisheng decoction group showed no significantly difference compared with Sodium hyaluronate group（ P〈0. 05）. Conclusion Duhuo Jisheng decoction can decrease the joint synovial fluid levels of IL-1β and TNF-a,it can also protect Collagen II,which may contribute the thereutic effect of Duhuo Jisheng decoction on knee osteoarthritis.
