阳和汤对乳腺癌MDA-MB-231细胞NF-κB、IL-8表达的影响

摘　　要：目的:观察阳和汤含药血清作用于乳腺癌MDA-MB-231细胞后,核因子(Nuclear Factor,NF)-κB、白介素(Interleukin,IL)-8的变化,了解阳和汤含药血清对乳腺癌细胞活性与分泌能力的影响。方法:用不同药物剂量(4.5g/kg、9g/kg、18g/kg)制备的含药血清分别干预乳腺癌MDA-MD-231细胞,在24小时、48小时、72小时、96小时,用MTT法检测细胞生长情况。在24小时、48小时、72小时,用Western blot检测NF-κB蛋白表达情况。在24小时、48小时,用ELISA检测IL-8的分泌量。结果:MTT法检测表明随着含药血清药物剂量增加,OD值增加,细胞活力降低,差异有统计学意义(P<0.05)。Western blot结果显示,随着含药血清药物剂量的增加,NF-κB的表达逐渐降低,随着含药血清作用时间的延长,NF-κB的表达也逐渐降低,差异有统计学意义(P<0.05)。ELISA检测显示,随着含药血清药物剂量的增加,细胞IL-8分泌量减少,差异有统计学意义(P<0.05)。结论:阳和汤抑制MDAMB-231细胞的增殖,下调NF-κB的表达、减少IL-8的分泌。Objective： The changes of NF-KB and IL-8 and the influence on activity, secretory ability were examined when MDA-MB-231 cells of breast cancer were objected to serum with Yanghe Decoction. Methods： Serum with different drug dosages （4. 5 g/kg, 9 g/kg, 18 g/kg） intervened in MDA-MB-231 cells of breast cancer respectively. The growth of the cells was tested by MTr at 24h, 48h, 72h, 96h. The expression of proteins was detected by western blot at 24h, 48h, 72h. The secretion of IL- 8 was measured by ELISA at 24h, 48h. Results： With the dosages increasing, MTT indicated that the OD increased, which was more significant than that in the blank controlled group. With the increasing of serum with drugs, the expression of NF-KB de- creased gradually, and with the time extension, the expression of NF-KB decreased, which were statistically lower than those of the blank controlled group. With the increasing of serum with drugs, the secretion of IL-8 decreased, which was statistically lower than the blank controlled group. Conclusion ： Yanghe Decoction can inhibit the growth of MDA-MB-231 cells, and degrade the ex- pression of NF-KB and the secretion of IL-8.
