摘 要:目的:观察小檗碱对糖尿病大鼠肠系膜血管病变的干预作用。方法:24只SD大鼠随机分为对照组、模型组和小檗碱组,每组8只。模型组和小檗碱组腹腔注射链脲佐菌素(STZ,55mg/kg)制备糖尿病大鼠模型,对照组腹腔注射等量生理盐水。大鼠成模两周后,小檗碱组灌胃给予小檗碱(200mg/kg),对照组和模型组灌胃等量双蒸水,三组均每日一次,连续2周,比较各组体重及血糖变化。之后处死各组大鼠,取其肠系膜、各级血管及其周围组织制备石蜡切片,HE染色,显微镜观察各组大鼠肠系膜各级血管管径及血管和周围组织的病理学变化。结果:与对照组比较,模型组大鼠实验28天后体重减轻(P<0.01),血糖升高(P<0.01)。二、三级动静脉管径较对照组明显增大(P<0.05或P<0.01);血管弹性膜分层不清,内膜白细胞黏附增多;血管周围脂肪组织中脂肪细胞皱缩,大量炎性细胞浸润,神经节肿胀。小檗碱组较模型组体重、血糖无明显差异(P>0.05),但三级动脉管径和二级静脉管径较模型组明显收缩(P<0.05或P<0.01);而且小檗碱组较模型组血管弹性膜分层清晰,内膜白细胞黏附减少;脂肪细胞形态、炎性细胞浸润及神经节形态均有一定程度的改善。结论:小檗碱对糖尿病大鼠肠系膜小血管和微血管及周围组织的病理变化有一定改善作用。Objective:To observe the intervention effect of berberine(BBR)on diabetic rat mesenteric vascular lesion.Method:24SD rats were divided into normal group,diabetic model(DM)group and BBR group randomly(n=8).Rats in DM group and BBR group were injected streptozotocin(STZ)(55mg/kg)to induce diabetic rat model,the normal group rats were injected with the same amount of saline.After two weeks of STZ injection,BBR group rats were administrated BBR(200mg/kg)by gavage,normal and DM group rats were administrated equal amount of distilled water once a day for two weeks.Compared the changes of body weight and blood glucose,and then killed all of rats and took the vascular tissues for routine treatment.Paraffin sections of vascular tissues were made.After HE staining,the circumference of vessel and histopathological changes of the mesenteric vascular at all levels and peripheral tissues were observed by microscopy.Results:In DM group rats,body weight was reduced(P〈0.01),level of blood glucose was increased significantly(P〈0.01),compared with the normal group after 28 days.The circumferences of the secondary and tertiary arteries(veins)were significantly increased,compared with the normal group(P〈0.05,P〈0.01).In DM group,the elastic membrane layer was unclear,endometrial leukocyte adhesion was increased,fat cells in perivascular adipose tissue shrinked,a large number of inflammatory cells infiltrated,and ganglion swelling.The BBR group had no significant difference in body weight and blood glucose compared with the control group(P〉0.05),but the circumferences of the tertiary artery and secondary vein were decreased compared with the DM group significantly(P〈0.05 or P〈0.01).After BBR treated,the vascular elastic membrane layer was clear,endometrial leukocyte adhesion was decreased.Adipocytes morphology,inflammatory cell infiltration and ganglion morphology was improved to a certain extent.Conclusion:BBR improves the small vessels,microvascular and surrounding
