电针百会、足三里穴对脑缺血再灌注大鼠凋亡相关因子Bcl-2和Bax表达的影响

摘　　要：目的:观察电针百会、足三里穴对脑缺血大鼠Bcl-2和Bax表达的影响,从细胞凋亡的角度探讨电针治疗该病的机制。方法:SD大鼠随机分为假手术组、模型组和电针组,根据再灌注时间又分为24 h和72 h组。假手术组:仅分离血管,不插入线栓,固定在布袋20 min,不进行电针治疗。模型组:制作脑缺血再灌注模型,每日固定同假手术组。电针组:电针百会穴和左侧足三里穴,日1次,留针20 min。规定时间点取大鼠脑组织,以免疫蛋白印记法及实时荧光定量PCR法检测缺血脑组织Bcl-2、Bax蛋白及mRNA表达。结果:电针组Bcl-2蛋白及mRNA表达显著高于同一时间点的模型组(P<0.05),Bax蛋白及mRNA表达显著低于同一时间点的模型组(P<0.05)。结论:电针大鼠百会、足三里穴能上调Bcl-2蛋白及基因表达水平,下调Bax蛋白及基因水平,电针减轻缺血再灌注后脑损伤,可能与上述调节相关。Objective：To observe the expression levels of Bcl -2 and Bax after treating acute cerebral infarction of rats with eletro - acupuncture so as to investigate the underlying mechanism of electro - acupuncture on ischemic stroke from the angle of apoptosis. Methods ： Healthy male Sprague - Dawley rats were randomly divid- ed into a sham operation （S） group, a model（M） group, an electro- acupuncture （EA） group, and then they were divided into 24h and 72h reperfusion subgroup. The S group and the M groups were bound by a bag for 20min as the rats in EA group. The rats in M group and EA group were made middle carotid artery occlusion reperfusion models. The rats in EA group were treated with electro - acupuncture treatment daily, and GV20 and left ST36 were selected. The protain levels of Bcl -2 and Bax in brain were evaluated by western blot and the mRNA levels of Bcl - 2 and Bax in brain were evaluated by Real - time PCR. Results： Compared with the M group, the protein and mRNA levels of Bcl - 2 in the EA group significantly increased at the same time point （P 〈 0.05）; Bax expressions in EA group were significantly lower than those in M group at the same time point. Conclusion： Electro- acupuncture at GV20 and ST36 can increase the protein and mRNA levels of Bel - 2, and decrease the protein and mRNA expression level of Bax in rat models of MCAO. Electro - acupuncture therapy has a protective effect on the focal CIRI in rats probably by regulating the expressions of Bcl - 2 and Bax in the apoptosis process.
