摘  要:目的探讨辽宁省与日本流行北京基因型结核分枝杆菌基因的异同。方法选取辽宁省各市结核病防治所经菌种鉴定得到的144株结核分枝杆菌菌株,以及84株来自日本神奈川县卫生研究所的结核分枝杆菌菌株,采用以聚合酶链式反应(PCR)为基础的分型方法鉴定是否为北京基因型结核分枝杆菌。对引物4扩增辽宁省北京基因型和非北京基因型结核分枝杆菌的条带进行双向测序并比较。结果 144株辽宁省结核分枝杆菌中,北京基因型结核分枝杆菌129株(89.58%),非北京基因型结核分枝杆菌7株(4.86%),北京基因型结核分枝杆菌和非北京基因型结核分枝杆菌混合感染8株(5.56%)。引物4扩增辽宁省7株非北京基因型结核分枝杆菌菌株得到308bp条带,扩增129株辽宁省北京基因型结核分枝杆菌菌株,116株(89.92%)北京基因型结核分枝杆菌出现270 bp左右条带。随机对3株辽宁省北京基因型结核分枝杆菌的270 bp左右条带进行双向测序,序列一致,与非北京基因型结核分枝杆菌的308 bp条带序列不同。84株日本神奈川县的结核分枝杆菌菌株中,北京基因型结核分枝杆菌53株(63.10%),非北京基因型结核分枝杆菌31株(36.90%),引物4扩增日本神奈川非北京基因型结核分枝杆菌菌株得到308 bp条带,扩增北京基因型结核分枝杆菌无条带。结论辽宁省以北京基因型结核分枝杆菌菌株流行为主,存在北京基因型和非北京基因型结核分枝杆菌混合感染。辽宁省流行北京基因型结核分枝杆菌的基因与日本流行的北京基因型结核分枝杆菌存在着不同的特点。Objective To discuss the similarities and differences of the gene of Beijing genotype M. tuberculosis in Liaoning Province and in Japan. Methods PCR method was used to identify whether 144 clinical isolated Mycobacterium tuberculosis in Liaoning province and 84 Mycobacterium tuberculosis at the Kanagawa in Japan were Beijing genotype Mycobacterium tuberculosis or not. The DNA Bands amplified M. tuberculosis in Liaoning Province by primer 4 were sequenced and compared. Results In the 144 cases in liaoning province,Beijing genotype mycobacterium tuberculosis were 129 cases( 89. 58%). Non-Beijing genotype M. tuberculosis was 7 cases( 4. 86%). Beijing and non-Beijing genetype Mycobacterium tuberculosis mixed infection were 8 cases( 5. 56%),when primer 4 was used to amplify non-Beijing genotype M.tuberculosis,get the 308 bp bands. On the other hand,when primer 4 was used to amplify Beijing genotype M. tuberculosis in Liaoning province,there were many bands. The 270 bp band appear when 116 Beijing genotype Mycobacterium tuberculosis were amplified by the use of primer 4. The three 270 bp band of the 116 bands were sequenced in bidirectional sequencing. The sequencing results of the three 270 bp bands were same,while they were different from the sequence of308 bp bands. In the 84 cases at the Kanagawa in Japan,Beijing genotype mycobacterium tuberculosis were 53 cases( 63. 10%). Non-Beijing genotype M. tuberculosis was 31 cases( 36. 90%). When Primer 4 was used to amplify Beijing genotype M. tuberculosis at the Kanagawa in Japan,there was not the band. When primer 4 was used to amplify non-Beijing genotype M. tuberculosis,could get the 308 bp bands. Conclusion Beijing genotype Mycobacterium tuberculosis are the main popular in Liaoning Province. There are some multiple infections in the cases in Liaoning Province. There are the differences between the gene of Beijing genotype M. tuberculosis in Liaoning Province and that in Japan.
