雪灵芝粗多糖对环磷酰胺致免疫抑制小鼠免疫功能的影响

摘　　要：目的探讨雪灵芝粗多糖(arenaria kansuensis crude polysaccharide,AKCP)对环磷酰胺致免疫抑制小鼠免疫功能的影响。方法将小鼠随机分为5组:空白对照组、环磷酰胺(CTX)模型组、AKCP高、中、低剂量组。除空白对照组外其他4组采用腹腔注射CTX建立小鼠免疫低下模型。各组分别给予蒸馏水、不同剂量AKCP灌胃,10 d后处死小鼠,检测体质量增率、胸腺指数、脾脏指数、脾淋巴细胞增殖活性和CD3^+/CD4^+/CD8^+亚群、NK细胞杀伤活性、腹腔巨噬细胞吞噬鸡红细胞能力、血清中IL-2和IFN-γ水平。结果 CTX组除NK细胞杀伤活性外的上述各项指标均较对照组明显降低(P<0.05),不同剂量AKCP作用可减轻CTX所致的各项指标下降,其中AKCP高、中剂量组体质量增率、胸腺指数、脾淋巴细胞CD3^+及CD3^+CD8^+亚群、巨噬细胞吞噬率与吞噬指数均较CTX模型组明显升高(P<0.05),AKCP低剂量组脾脏指数、脾淋巴细胞增殖指数以及IL-2和IFN-γ水平显著高于CTX模型组(P<0.05)。结论 AKCP对环磷酰胺致免疫抑制小鼠的免疫功能有保护作用。This study aimed to explore the effects of crude polysaccharide from Arenaria kansuensis（AKCP）on immune function of immunosuppression mice induced by cyclophosphamide. Mice were randomly divided into five groups： control group, cyclophosphamide（CTX） group and three AKCP in high, middle and low-dosages groups.Except control group, other groups＇ mice received CTX i.p. to establish immunosuppressive model. After AKCP were administrated by i.g. for 10 days, the mice were killed, and the weight gain rate, the thymus and spleen index,the spleen lymphocyte proliferation as well as CD3^＋/CD4^＋/CD8^＋ subsets, NK killing activity, phagocytosis activity ofperitoneal macrophages, the levels of IL-2 and IFN-γ in serum were detected. Data showed that except NK killingactivity, other items in CTX group were all significantly decreased compared with control group（P〈0.05）. AKCP ofdifferent-dosage treatment could relieve those effects. In high and middle-dose AKCP groups, the weight gain rate,thymus index, CD3^＋ and CD3^＋CD8^＋T cells in spleen, phagocytosis activity of macrophage were significantly higherthan those in CTX group（P〈0.05）; in low-dose AKCP group, the spleen index, lymphocyte proliferation index, and the levels of IL-2 and IFN-γ in serum were obviously higher than those in CTX group（P〈0.05）. In conclusion,AKCP could protect immune functions of immunosuppression model mice induced by CTX.
