摘  要:采用PCR扩增、测序分析、接合试验、药敏试验、脉冲场凝胶电泳(PFGE)和Southern blot方法,分析了1株兽医临床分离大肠杆菌(sd-3)携带的超广谱β-内酰胺酶(ESBL)基因的状况、分型、定位及遗传环境。结果显示,该株大肠杆菌携带ESBL基因,属于bla_(CTX-M-9)亚群的Bla_(CTX-M-125)亚型,系在大肠杆菌中首次检出;接合试验、PFGE和Southern blot证实该基因位于接合型质粒上;Bla_(CTX-M-125)基因的上、下游分别存在插入序列ISEcp1和IS903。这表明插入序列和接合型质粒均可在大肠杆菌Bla_(CTX-M-125)基因的水平转移中发挥重要作用,有必要在肠杆菌科细菌中开展该基因的监视。The presence,types,location and genetic environment of extended-spectrum β-lactamases( ESBLs) gene in a Escherichia coli strain( sd-3) isolated from veterinary clinic were investigated by PCR amplification,sequencing analysis,conjugation assay,antimicrobial susceptibility test,pulsed field gel electrophoresis( PFGE) and Southern blot. The results showed the strain carried Bla_(CTX-M-125) ESBLs,belonging to the blaCTX-M-9subgroup,which was firstly described in Escherichia coli. Conjugation,PFGE and Southern blot confirmed that the gene was located on the conjugative plasmid. The upstream and downstream of the Bla_(CTX-M-125) was flanked by insertion sequence ISEcp1 and IS903,respectively. This indicated that both insertion sequence and conjugative plasmid had played an important role in the horizontal transfer of the Bla_(CTX-M-125) ESBL gene. The surveillance of the Bla_(CTX-M-125) in enterobacteriaceae bacteria is necessary.
