摘 要:目的研究石菖蒲挥发油有效成分联合左旋多巴对6-羟基多巴诱导帕金森病模型大鼠血脑屏障的影响。方法定位于SPF级SD大鼠左脑的内侧前脑束(前囟后:-4.4 mm,中线旁:-1.2 mm,硬膜下:-7.8 mm),注射24μg的6-羟基多巴,造模1个月后,将造模大鼠腹腔注射0.5 mg/kg的阿扑吗啡,大鼠向健侧旋转数大于7圈/min为成功PD模型。将40只PD模型大鼠随机分为4组,每组10只,雌雄各半,即模型组,左旋多巴组(60 mg/kg),β-细辛醚组(15 mg/kg),联合用药组(左旋多巴60 mg/kg+β-细辛醚15 mg/kg)。给药组按上述剂量灌胃给药,一天2次,给药30天。末次给药1 h后,冰上迅速分离皮质。高效液相色谱法(HPLC)检测左旋多巴和多巴胺含量;流式细胞术检测皮质紧密连接蛋白(Zonula occludens 1,ZO-1)、紧密连接相关蛋白(Occludin)和肌动蛋白(Actin)含量;免疫组化检测紧密连接相关蛋白5(Claudin-5)。结果与假手术组比较,模型组大鼠的左旋多巴和多巴胺含量都显著减少(P<0.05),ZO-1、Occludin、Actin和Claudin-5的表达无显著变化(P>0.05);与模型组比较,联合用药组大鼠的左旋多巴和多巴胺含量都显著增加(P<0.01),ZO-1、Occludin、Actin和Claudin-5的表达都显著减少(P<0.05);与左旋多巴组比较,联合用药组大鼠的左旋多巴和多巴胺含量显著增加(P<0.05),联合用药组大鼠的ZO-1、Occludin、Actin和Claudin-5的表达都显著减少(P<0.05)。结论石菖蒲挥发油有效成分β-细辛醚联合左旋多巴具有一定提高PD模型大鼠皮质中BBB通透性的作用。Objective To study the effect of the effective components of volatile oil of Acorus atarinowii Schott and levodopa( L-dopa) co-administration on blood brain barrier in 6-hydroxydopamine induced Parkinson’s rat. Methods Twenty-four μg 6-OHDA was then injected into medial forebrain bundle( MFB) of each rat at the following: antero-posterior:-4. 4 mm,medio-lateral:-1. 2 mm,dorso-ventral:-7. 8 mm. One month after the injection,seventy PD rats were successfully chosen according to apomorphine( 0. 5 mg/kg,i. p.) ratation test( 7 circles/min). The PD rats were divided into four groups( n = 10) : model group,L-dopa group,β-asarone group,L-dopa + β-asarone group. All the treated-rats were given by intragastric administration( 5 m L/kg) twice a day for consecutive 30 days. Then levodopa and dopamine levels were determined by high performance liquid chromatography( HPLC). Zonula occludens-1( ZO-1),claudin-5,occludin and actin expressions in prefrontal cortex were detected by flow cytometry. Claudin-5 expression was detected by immunohistochemistry. Results Compared with control group,model group rats levodopa and dopamine levels were significantly reduced( P 〈 0. 05),the expressions of ZO-1,Occludin,Actin and Claudin-5 had no significant change( P 〉 0. 05). Compared with model group,combination group rats levodopa and dopamine levels were significantly increased( P 〈 0. 01),the expression of ZO-1,Occludin,Actin and Claudin-5 were significantly reduced( P 〈 0. 05). Compared with levodopa group,combination group rats levodopa and dopamine levels increased significantly( P 〈 0. 05),the expression of ZO-1,Occludin,Actin and Claudin-5 were significantly reduced( P 〈 0. 05). Conclusion Co-administration of β-asarone in effective components of volatile oil of Acorus tatarinowii Schott and levodopa has an effect on permeability of blood brain barrier in 6-hydroxydopamine induced Parkinson’s rat.
