摘 要:目的观察龟羚帕安丸对帕金森病(PD)大鼠中脑黑质IL-1β、IL-6、i NOS、IFN-γ表达的影响。方法采用6-OHDA注射法建立PD大鼠模型,造模成功大鼠随机分为模型组,美多芭组(10mg·kg-1),龟羚帕安丸高、中、低剂量组(4,2,1g·kg-1),同时设立假手术组。假手术组、模型组给予等容积生理盐水,其他组予相应药物,连续灌胃给药30天。免疫组织化学法检测中脑黑质IL-1β、IL-6、i NOS、IFN-γ蛋白表达。结果龟羚帕安丸能明显抑制炎症因子IL-1β、IL-6、i NOS、IFN-γ表达。结论龟羚帕安丸通过降低IL-1β、IL-6、i NOS、IFN-γ表达,通过抗炎作用,对多巴胺能神经元产生保护作用,可能是龟羚帕安丸治疗PD的作用机制之一。Objective To investigate the effect of Guiling Pa’an pill on the expression of IL-1β,iNOS,IL-6 and IFN-γin substantia nigra of rats with Parkinson’s disease( PD). Methods The rats were stereotaxically injected with 6-OHDA solution into the left striatum. The rats was judged as PD model. Fifty model rats were randomly divided into five groups: model group,madopar group( 10mg·kg-1),Guiling Pa’an pill group( 1. 0,2. 0,4. 0g·kg-1),at the same time,the sham operation group was established and given normal saline at the same volume,all with a course of 30 days. The IL-1β,i NOS,IL-6 and IFN-γ protein levels of the substantia nigra tissue were detected by immunohistochemistry. Results Compared with sham group,the IL-1β、i NOS、IL-6 and IFN-γprotein in substantia nigra tissue of model group were significantly increased( P 〈 0. 01). Compared with model group,the IL-1β,i NOS,IL-6 and IFN-γ protein in substantia nigra tissue were significantly decreased( P 〈0. 01). Conclusion The mechanism of efficacy of Guiling pa’an pill in the treatment of PD by reducing the IL-1β,i NOS,IL-6and IFN-γ expression,the anti-inflammatory effects of dopaminergic neurons,have a protective effect.
