摘 要:目的通过研究钙对染铅大鼠海马NMDAR和m Glu R5受体介导相关基因mRNA及CaMKII蛋白表达的影响,为进一步以补钙预防铅的危害提供理论依据。方法选择体质量(50±2)g的清洁级雄性SD大鼠60只,共分五组,每组12只,分别为对照组(基础饲料,蒸馏水)、染铅组(基础饲料,0.2%醋酸铅饮水)、试验Ⅰ组(基础饲料添加0.5%碳酸钙,0.2%醋酸铅饮水)、试验Ⅱ组(基础饲料添加1%碳酸钙,0.2%醋酸铅饮水)和试验Ⅲ组(基础饲料添加2%碳酸钙,0.2%醋酸铅饮水)。连续饲养60 d后,取海马,用荧光定量RT-PCR法测定海马中Grm5、Camk2a、Grin1、Homer1、Prkca和ERK2的mRNA表达量,Western-blotting测定大鼠海马中CaMKII的蛋白表达量。结果与对照组相比,染铅组海马中Grm5、Camk2a、Grin1、Homer1、Prkca、ERK2的mRNA表达量及CaMKII的蛋白表达量均极显著降低(P<0.01)。与染铅组相比,试验组钙浓度在一定范围内越高,目的基因表达量相对升高越多,试验Ⅱ组CaMKII的蛋白表达量极显著上升(P<0.01)。结论一定浓度的钙可干预铅致大鼠海马组织的损伤。Objectives To investigate the effect of calcium on the Mrna expression of the genes related to NMDAR or mGluR5 and the protein expression of CaMKII of hippocampus in rats exposed to lead, and provide theoretical basis for the prevention of the hazard induced by lead with calcium supplement.Methods Sixty healthy male rats(50±2)g were randomly divided into five groups, normal control group (normal diet, distilled water), lead exposed group (normal diet, 0.2% lead acetate water), testⅠgroup (0.5% calcium carbonate + normal diet, 0.2% lead acetate water), testⅡgroup (1% calcium carbonate + normal diet, 0.2% lead acetate water) and testⅢ group (2% calcium carbonate + normal diet, 0.2% lead acetate water). After 60 days of continuous feeding, the hippocampus were collected. The Mrna expression levels of Grm5, Camk2a, Grin1, Homer1, Prkca and ERK2 were measured by real-time RT-PCR. In addition, CaMKII protein expression in rat hippocampus was determined by Western-blotting.Results Compared with the normal control group, the Mrna expression levels of Grm5, Camk2a, Grin1, Homer1, Prkca, ERK2 and the CaMKII protein in the hippocampus of the rats exposed to lead were significantly reduced. When compared with the rats exposed to lead without calcium supplement, their expression levels increased in the rats of three test groups, and the increase of CaMKII protein expression in test group II was statistically significant .Conclusion These Results suggest that calcium can protect against the damage of hippocampus induced by lead exposure.
