Effects of Jianpiyizhi Capsule on Expression of NT3 in the Hippocampus and Learning and Memorizing Ability of MCAO Rats

摘  要:[目的]探讨健脾益智胶囊对大脑中动脉栓塞(MCAO)大鼠海马神经营养素3(NT3)及学习记忆能力的影响。[方法]建立MCAO大鼠模型,随机分为正常组、假手术组、模型组、西药组、中药组。采用免疫组化法、Bederson评分、Morris水迷宫实验、跳台实验于术后7 d、14 d、28 d观察MCAO大鼠海马NT3的表达及学习记忆能力的改善情况。[结果]随着术后时间的增加,模型组、西药组、中药组大鼠Bederson评分逐渐减少,组内各时间点的评分均有显著性差异(P<0.05);与模型组比较中药组与西药组同时间点评分差异显著(P<0.05),且中药组评分优于西药组(P<0.05)。随着术后时间的增加,Morris水迷宫实验中模型组、中药组、西药组逃避潜伏期逐渐缩短,穿越原平台次数逐渐增多;跳台实验潜伏期逐渐延长,错误次数逐渐减少(P<0.05)。中药组均较同时间点模型组、西药组水迷宫实验逃避潜伏期缩短,穿越次数增多;跳台实验潜伏期延长,错误次数减少(P<0.05)。免疫组化实验显示,术后7 d中药组NT3阳性细胞数明显多于模型组、西药组(P<0.05);术后14 d中药组NT3阳性细胞数与西药组无明显差异(P>0.05);术后28 d中药组NT3阳性细胞数明显多于模型组、西药组(P<0.05)。[结论]健脾益智胶囊可促进MCAO大鼠学习记忆能力的恢复,促进MCAO大鼠海马NT3表达增加,提示健脾益智胶囊可能通过诱导NT3表达以达到恢复脑缺血后学习记忆能力的目的。[ Objective ] To detect the effects of Jianpiyizhi capsule on learning and memorizing ability of middle cerebral artery occlusion (MCAO) rats. [Methods] The MCAO model rats were randomly divided into 5 groups: normal control group, sham operation group, MCAO model group, Nimodipine group, Jianpiyizhi group. The expression of NT3 in hippocampus and the learning and memory ability of MCAO rats were observed by immunohistochemical method, Bederson score, Morris water maze test and step- down test at 7d, 14d and 28d after operation. [Results]With the increase of the postoperative time, the Bederson scores of MCAO model group, Nimodipine and Jianpiyizhi group gradually reduced. Bederson scores were significant different in each group at each time point (P〈O.05). The scores of Nimodipine and Jianpiyizhi groups at the same time-point were significant different compared with that of MCAO model group (P〈0.05). And the score of Jianpiyizhi group was better than that of Nimodipine group (P〈0.05). With the increase of the postoperative time, the escape latency of MCAO model group, Nimodipine and Jianpiyizhi groups was gradually shortened and the frequency of crossing the original platform was gradually increased in Morris water maze test. The latency of the experiment was gradually prolonged and the number of errors decreased gradually (P〈0.05). At the same time point, the escape latency, frequency of crossing the original platform, the latency of the experiment and the number of errors in Morris water maze test of Janpiyizhi group was significantly differently from that of MCAO model group and Nimodipine group (P 〈0.05). Immunohistochemical experiments showed that the number of NT3 positive cells on the 7th day after surgery of the Jianpiyizhi group were significantly higher than that in MCAO model group and Nimodipine groups(P〈0.05). No significant difference was noted in the number of NT3 positive cells on the 14th day after surgery between Janpiyizhi group and Nimodipine group(
