摘 要:宫颈癌是常见的女性恶性肿瘤,其发生发展与HPV的感染密切相关。HPV16是宫颈癌患者中感染率最高的亚型,其编码的早期蛋白E5、E6、E7可刺激宫颈上皮细胞转化成肿瘤细胞。E6、E7蛋白在宫颈癌晚期表达,E5可能在早期表达。E5蛋白的是否表达及其致癌机制与多种因子相关。E5蛋白可作用于细胞生长因子受体通路,下调miRNA196a,并通过与HLAⅠ类分子结合协助感染细胞免疫逃逸,从而促进肿瘤的发生。E5蛋白与E6、E7蛋白协同会发挥更强的致宫颈癌作用。本研究对E5蛋白表达与病毒整合的关系及其对宫颈癌的作用进行了综述。Cervical cancer is a common malignant tumor in women and HPV is closely related with the occurrence and development of cervical cancer. Human papillomavirus type 16( HPV16) is the highest subtypes of HPV infection in cervical carcinoma patients and the E5、E6 and E7 oncoproteins encoded by HPV16 could translate cervical epithelial cells into tumor cells. E6 and E7 gene express in advanced cervical carcinoma,but E5 gene may express in the early stage of the cervical cancer. E5 protein can increase the occurrence of tumorigenesis by regulating epidermal growth factor receptor( EGFR) signaling pathways,reducing miRNA 196 a,and interacting with HLA class to help the infected cells to immune evasion. E5 protein can also cooperate with E6 protein and E7 protein to play a greater role in tumorigenesis of cervical cancer. The relationship between the expression of E5 protein and virus integration and the role of E5 protein in cervical cancer are reviewed in this article.
