摘 要:目的:研究人参皂甙Rh2对H_2O_2损伤保护作用及与GABAA受体的关系。方法:采用小鼠制备离体海马脑片,随机分为正常组、模型组、GABA组、人参皂甙Rh2(20μmol/L、40μmol/L、60μmol/L)组、荷包牡丹碱组、荷包牡丹碱+人参皂甙Rh2组,每组20张脑片。采用碘化丙啶(PI)染色法测定H_2O_2损伤后海马CA1区细胞活力变化,免疫组化方法观察细胞色素C表达,电生理技术记录顺向群峰电位(OPS)观察突触传递变化。结果:人参皂甙Rh2(40μmol/L、60μmol/L)可明显提高H_2O_2损伤后海马CA1区细胞活力,降低海马CA1区细胞色素C表达,海马脑片OPS恢复率和OPS恢复程度亦明显提高。人参皂甙Rh2的保护作用和外源性GABA作用相似并可被GABAA受体抑制剂荷包牡丹碱所阻断。结论:人参皂甙Rh2可减轻H_2O_2对海马CA1区细胞损伤,促进神经元突触传递功能的恢复,其作用机制与激活GABAA受体而减少细胞色素C的释放有关。Aim:To investigate the protective effect of ginsenoside Rh2 on H2O2 injury in hippocampal slices and therole of GABAA receptor.Methods:Mouse hippocampal slices were randomly divided into normal group, model group, GABA group, ginsenoside Rh2 (20μmol/L, 40μmol/L,60μ mol/L) group, bicuculline group and bicuculline + ginsenoside Rh2 group. The cellular viability after H2O2 injury was deter-mined by propidium iodide ( PI ) staining, the changes of orthodromic population spike ( OPS ) were observed by electrophysiological technique, the expressions of cytochrome C in hippocampal CA1 regions were measured by immunohistoehemical assay. Results:Ginsenoside Rh2 ( 40μmol/L and 60μmol/L) improved significantly the recovery rate and amplitude of OPS ,which also significantly increased the cellular viability and significantly decreased the expression of cytochrome C in hippocampal CA1 regions after H2O2 injury. The effect of ginsenoside Rh2 was similar to exogenous GABA and reduced by GABAA receptor antagonist bicuculline. Conclusion:Ginsenoside Rh2 can antagonize H2O2 injury and improve the recovery of synaptic transmissioni neurons. The mechanism is related to activation of GABAA receptor and reduction of cytochrome C release.
