摘 要:目的:观察青藤碱和美沙酮对斑马鱼胚胎的心脏毒性。方法:以受精后48 h(48 hours post-fertilization,48 hpf)发育正常的斑马鱼胚胎作为心脏毒性模型,用不同浓度的青藤碱(1、0.5、0.3、0.2、0.1 g/L)和美沙酮(1、0.5、0.3、0.2、0.1 g/L)处理上述胚胎,分别于处理后12 h和24 h观察胚胎的心脏形态及心率变化。结果:各剂量青藤碱组斑马鱼胚胎心率与正常对照组相比均无显著性差异,心脏区域均未出现淤血堆积、心包肿大等中毒现象。1、0.5 g/L美沙酮组斑马鱼胚胎在给药12 h后全部死亡;0.3、0.2g/L美沙酮组斑马鱼胚胎出现部分死亡,存活斑马鱼胚胎心率与正常对照组相比均显著降低,并出现血细胞在心区堆积、心包囊水肿及心膜出血等心脏中毒现象;0.1 g/L美沙酮组斑马鱼胚胎心率及心脏形态与正常对照组相比无显著差异。结论:青藤碱无明显的斑马鱼胚胎毒性,而美沙酮对斑马鱼胚胎有心脏毒性作用。Objective: To investigate the eardiotoxieity of sinomenine and methadone in zebrafish embryos. Methods: After 48 hours post-fertilization (hpf), zebrafish embryos were randomly divided into sinomenine groups at doses of 1,0.5,0.3,0.2,0.1 g/L, methadone groups at doses of 1,0.5,0.3,0.2,0.1 g/L and control group. At 12 h and 24 h after administration,the pericardium and cardiac morphology of zebrafish em- bryos were observed by microscope and the heart rate was recorded. Results:There were no significant differences in the heart rates of zebrafish embryos in all sinomenine in comparison with that of control animals and neither cardiac congestion nor enlargement of pericardium appeared after 12 h and 24 h of administration. After exposured to methadone, all zebrafish embryos died at 12 h after administration in the doses of 1,0.5 g/L,while embryos were partly dead in groups in the doses of 0.3,0.2 g/L and the heart rates of the survivors were significantly reduced in eomparision with that of control animals. The phenomena of cardiotoxieity such as cardiac congestion and enlargement and bleeding of pericardium appeared in zebrafish embryos. No significant difference of the heart rate and cardiac morphology of embryos were observed be- tween the 0.1 g/L methadone and control group. Conclusion: Sinomenine didnt have obvious cardiotoxicity and methadone had cardiotoxic effect in zebrafish embryos.
