摘  要:目的:筛选火炭母治疗乳腺增生的活性部位。方法:用戊酸雌二醇配合黄体酮成功建立乳腺增生小鼠模型,将180只小鼠分为正常对照组、模型对照组、乳癖消贴膏对照组、给药组(5个提取分离部位)。通过将火炭母的各提取分离部位分别制成软膏,每日外用给药1次,连续给药7天。用游标卡尺测量小鼠乳房直径、乳头高度;末次给药后眼底取血,ELISA法测定血清雌二醇水平;分析天平称量子宫、卵巢重量,测定子宫卵巢指数;取第二对乳腺进行病理学检查。结果:火炭母乙醇提取物的各部位均可以降低小鼠乳房直径和乳头高度,降低雌激素水平,减轻乳腺组织增生,其中氯仿部位(给药剂量21、42 mg/kg)、甲醇部位(给药剂量358、716mg/kg)效果更明显。结论:火炭母氯仿部位、甲醇部位对小鼠乳腺增生有显著的治疗作用,为火炭母的临床应用提供科学依据,为进一步研究火炭母的化学成分及药理作用奠定基础。Objective: Screening the bioactive fractions of Polygonum chinense for the treatment of breast hyperplasia . Methods: Establishing breast hyperplasia model of mice by administration of estradiol valerate and progesterone, bioactive screening on five fractions of Polygonum chinense was performed. After administration of the ointment made of different fractions of P. chinense respectively for 7 d, parameters such as breast diameter, nipple height, uterus weight, ovary weight and serum estradiol levels of the mice were measured to determine the bioactive fractions of P. chinense for the treatment of mammary gland hyperplasia. Results: each part of ethanol extract of P. chinense can decrease mouse breast diameter( P 〈 0.05 ) and breast height( P 〈 0.01 ) , lower the level of estrogen( P 〈 0.01 ) , relieve breast hyperplasia, wherein fractions of chloroform or methanol showed more potent effect. Conclusion : The results of this paper can not only provide scientific accordings for clinical employment of P. chinense, but lay a reliable foundation for further investigation of chemical constituents and pharmacological effect on this plant.
