洋金花总碱对人肺腺癌A549细胞EMT过程中E-cad表达的影响

摘  要:目的观察洋金花总碱对转化生长因子-β1(TGF-β1)诱导的A549细胞向间质细胞转化(EMT)过程中E-钙黏蛋白(E-cad)表达的影响。方法将体外培养的A549细胞随机分为空白对照组,TGF-β1组及洋金花总碱低、中、高浓度药物组,分别作用24、48、72h后,通过Western-blot及RT-PCT检测各组A549细胞E-cad表达的变化。结果 24、48、72h E-cad表达,空白对照组分别为(1.49±0.12)、(2.52±0.07)、(2.58±0.21);TGF-β1组分别为(0.66±0.01)、(0.50±0.13)、(0.52±0.08);洋金花总碱低、中、高浓度药物组分别为24h[(0.67±0.12)、(0.67±0.12)、(1.12±0.05)]、48h[(0.70±0.28)、(1.02±0.08)、(1.10±0.13)]、72h[(0.68±0.04)、(1.25±0.11)、(1.30±0.09)]。与空白对照组比较,TGF-β1组和洋金花总碱低、中、高浓度药物组的E-cad表达均明显减少(P<0.01);与TGF-β1组比较,各时间段的洋金花总碱低浓度组E-cad表达无明显变化(P>0.05),但48、72h洋金花总碱中、高浓度组E-cad表达明显增加(P<0.01);与24h低浓度药物组比较,24h中浓度药物组Ecad表达差异无统计学意义(P>0.05),但作用48、72h后,中浓度药物组的E-cad表达明显增加(P<0.01),而高浓度药物组3个时间点的E-cad表达均明显增加(P<0.01)。与中浓度药物组比较,24h高浓度药物组E-cad表达增加(P<0.05),48、72h高浓度药物组E-cad表达差异无统计学意义(P>0.05)。与空白对照组比较,不同浓度作用的洋金花总碱药物组使E-cad m RNA表达上调(P<0.01),并且在72h及高浓度时最明显(P<0.01)。结论洋金花总碱通过增加上皮标志物E-cad表达,可抑制A549细胞的上皮间质转化。Objective To observe the E-cad expression changes in A549 cells during TGF-β1-induced EMT after Yangjinhua alkaloids treatment. Methods A549 cells in vitro cultured were randomly divided into blank control group, TGF-β1 group, low, medium and high dose of Yangjinhua alkaloids group. The change of cell morphology and the expression of E-cad were observed by western blot and RT-PCR after 24, 48 and 72 hours, respectively. Results The expressions of E-cad at 24, 48 and 72 hours after treatment were 1.49 ±0.12, 2.52 ±0.07,2.58±0.21 in blank control group, 0.66±0.01, 0.50±0.13, 0.52±0.08 in TGF-β1 group, 0.67±0.12, 0.67±0.12, 1.12±0.05 in low dose of Yangjinhua alkaloids group, 0.70±0.28, 1.02±0.08, 1.10±.13 in medium dose of Yangjinhua alkaloids group and 0.68 ±0.04, 1.25 ±0.11, 1.30 ±0.09 in high dose of Yangjinhua alkaloids group. Compared with those in blank control group, the expressions of E-cad in three Yangjinhua alkaloids groups were significantly decreased(P〈0.01). Compared to those in TGF-β1 group, the expressions of E-cad at 24, 48 and 72 hours in low dose of Yangjinhua alkaloids group were not different(P〈0.05), but the expressions of E-cad at 48 and 72 hours in medium and high doses of Yangjinhua alkaloids groups were obviously increased(P 0.01). Among three Yangjinhua alkaloids groups, the expression of E-cad at 24 h in low dose group did not differ from that in medium dose group, but the expression of E-cad at 48 and 72 hours in medium dose groups were significantly increased(P〈0.01), and the expression of E-cad in high dose group were higher at each time point(P〈0.01); compared to those in medium dose group, the expression of E-cad at 24 h in high dose group were raised(P〈0.05),but no significant difference was noted between two groups at 48 and 72 hours(P〈0.05).
