生髓育麟汤对Caspase-3以及Bcl-2/Bax在实验性少、弱精子大鼠生精细胞中表达的干预

摘　　要：目的探讨生髓育麟汤治疗少、弱精子症的作用机理。方法将60只大鼠按照常规方法饲养,1周后随机抽出10只编入正常对照组,另外50只予雷公藤多苷片20 mg/(kg·d)灌胃26d。造模完毕后随机抽取10只编入造模组处死,剩下40只按照体重随机分为4组:生髓育麟汤高、低剂量组、五子衍宗丸组以及不给予任何药物,只给予正常食物和水的观察对照组,每组各10只。实验结束后获取大鼠附睾精子进行分析,了解大鼠精子质量、生精细胞凋亡率、Caspase-3以及Bcl-2/Bax表达水平。结果生髓育麟汤各组的精子密度、前向运动精子比例、精子活动率明显增加;生髓育麟汤各组生精细胞凋亡率下降,Caspase-3以及Bax表达减少而Bcl-2表达增加,并且随着浓度的增加,效果更显著。结论生髓育麟汤可调节生精细胞基因表达,抑制细胞的凋亡,从而改善大鼠的精液质量。Objective To explore the effect mechanism of shengsui yulin decoction（ the decoction for short-term）. Methods Sixty rats were fed regularly. One week later,10 rats of them were randomized into a normal control group and 50 rats of them were lavaged with tripterygium glycosides,20 mg/kg per day,for 26 days. After modeling,10 rats of them were sacrificed and divided into the modeling group and the rest40 rats were randomized into 4 observation control groups according to body weight,i. e. the high-dose decoction group,the low-dose decoction group and wuzi yanzong wan group and the non-medication group in which normal diet and water were used,10 rats in each one. At the end of experiment,the sperms were collected for analysis,understanding sperm quality,apoptosis rate of germ cell,and the expressions of Caspase-3 and Bcl-2/Bax. Results The sperm density,the proportion of forward movement sperms and sperm active ratio were increased apparently in each decoction group. In each decoction group,the apoptosis rate of germ cell was reduced,the expressions of Caspase-3 and Bax were reduced and the Bcl-2 expression was increased. With the increase of the concentration,the effect was more significant. Conclusion Shengsui yulin decoction regulates the expression of spermatogenic cell and inhibits cellular apoptosis so that the seminal fluid quality is improved in the rats.
