摘  要:目的探讨除湿通痹方对急性痛风性关节炎炎症级联反应的作用。方法 SD大鼠随机分为空白组、模型组、秋水仙碱组,除湿通痹方高、中、低剂量组共6组,每组10只,于给药前1d和给药第6天,除空白组给予生理盐水外,其余5组复制急性痛风性关节炎模型,连续灌胃22d后处死采取标本取材,观察各组大鼠关节肿胀度、功能障碍指数和炎症指数、测定IL-1β、TNF-α浓度,检测caspase-1、ASC、NLRP3基因表达,并分析其关系。结果各给药组对急性痛风性关节炎大鼠活动障碍均有缓解作用;秋水仙碱组TNF-α表达水平显著低于模型组(P<0.05):秋水仙碱组及除湿通痹方高剂量组IL-1β、caspase-1、ASC、NLRP3表达水平显著低于模型组(P<0.05);caspase-1、ASC、NLRP3均与IL-1β存在相关性(P<0.05);TNF-α与ASC存在相关性(P<0.05)。结论秋水仙碱及除湿通痹方高剂量能通过抑制NLRP3炎性体的表达,从而抑制IL-1β相关的炎性过程,发挥其抗炎免疫作用。Objective To investigate the effect of Chushi Tongbi Decotion of removing dampness and resolving dampness on the cascade reaction of acute gouty arthritis. Methods 60 SD rats were randomly divided into 6 groups: blank group, model group, colchicine group, Chushi Tongbi high-dose group, middle-dose group and low-dose group, 10 rats were included in each group. Before rats were given by gavage one day and the sixth day, acute gouty arthritis model of was replicated by intra-articular injecting monosodlum urate crystal suspension inside the ankle joint and only bhnk group mice were injected by NS. Rats were gavaged once a day for 22 days and joint swelling, inflammation index and dysfunction index were observed. IL-1β and TNF-α expression levels in soft tissues of joint were detected by ELISA, caspase-1, ASC, NLRP3 expression levels in soft tissues of joint were detected by qPCR and the relationship between each other was analyzed. Results Dysfunction index of all the rats in the treating groups on the acute gouty arthritis was relieved. TNF-α of expression of colchicine group was significantly lower than that of model group (P〈0.05); IL-1β, caspase-1 ,ASC ,NLRP3 of expression of colchicine and Chushi Tongbi high-dose group was significantly lower than that of model group (P〈0.05); there were significant correlations between IL-1β and caspase-1, ASC, NLRP3; there were significant correlations between TNF-α and ASC (P〈0.05). Conclusion Colchicine group and Chushi Tongbi high-dose group can decrease inflammatory of IL-1β by decreasing expression levels of NLRP3, and then play its anti-inflammatory immune function.
