红鱼眼叶化学成分对人肝癌BEL-7404细胞株增殖的影响

摘　　要：目的:观察红鱼眼叶不同提取物及其化学成分对人肝癌BEL-7404细胞增殖的影响。方法:采用MTT法,观察红鱼眼叶不同提取物的含药血清对人肝癌BEL-7404细胞的体外抑制作用并从有效部位分离出单体成分,观察各化学成分对肝癌BEL-7404细胞的影响。结果:与20%空白血清对照组比较,红鱼眼叶乙酸乙酯提取物20%含药血清对人肝癌BEL-7404细胞有明显的体外抑制作用(P<0.05);红鱼眼叶乙酸乙酯提取物中没食子酸、柯里拉京、短叶苏木酚酸乙酯对人肝癌细胞BEL-7404的增殖具有明显抑制作用,其IC_(50)分别为46.81,32.40,41.15μg·m L^(-1),没食子酸乙酯对人肝癌细胞BEL-7404的增殖基本无抑制作用,其IC_(50)为-6.67μg·m L^(-1)。结论:红鱼眼叶乙酸乙酯提取物能抑制人肝癌BEL-7404细胞的增殖;从红鱼眼叶乙酸乙酯中分离的没食子酸、柯里拉京、短叶苏木酚酸乙酯对人肝癌细胞BEL-7404的增殖具有抑制作用。OBJECTIVE To observe the effects of different extracts of Phyllanthus reticulatus and the chemical constituents against BEL-7404 cell proliferation. METHODS MTT method was applied to investigate the effects of serum containing different extracts of Phyllanthus reticulatus on proliferation of human hepatoma cell line BEL-7404,and effects of 4 kinds of constituents isolated from the extracts in vitro against proliferation of human hepatoma cell line BEL-7404. RESULTS Compared to blank control,20%serum containing extracts from leave of Phyllanthus reticulatus showed obviously inhibitory activity against proliferation of human hepatoma cell line BEL-7404 in vitro（ P 0. 05）. The results showed that corilagin（ IC_（50）= 46. 81 μg·m L^（-1））,ethyl brevifolincarboxylate（ IC_（50）= 32. 40 μg·m L^（-1））,gallic acid（ IC_（50）= 41. 15 μg·m L^（-1）） inhibited proliferation of human hepatoma cell line BEL-7404,but ethyl gallate had no inhibiting effect against the proliferation. CONCLUSION Extracts from leaves of Phyllanthus reticulatus have inhibitory effects against proliferation of human hepatoma cell line BEL-7404. Corilagin,ethyl brevifolincarboxylate,gallic acid can inhibit the proliferation of human hepatoma cell line BEL-7404.
