摘 要:目的:通过兔耳增生性瘢痕(HS)局部注射人参皂苷Rb_1,观察其治疗效果及探讨相关作用机制。方法:随机取8只新西兰大耳白兔,每耳6处创面,建立兔耳增生性瘢痕模型,上皮化后局部注射人参皂苷Rb_1,每3天1次,共3次。设空白组、地塞米松阳性对照组、生理盐水对照组、人参皂苷Rb_1组。注射后第1、2、4、8周切除标本行HE染色观察瘢痕增生指数(SEI),qPCR检测转化生长因子β(TGF-β)及Ⅰ型胶原表达。结果:注射后第4、8周,地塞米松及人参皂苷Rb_1组较生理盐水组HS瘢痕增生改善明显,瘢痕增生指数降低(P<0.05),TGF-β1及Ⅰ型胶原mRNA表达降低(P<0.05)。结论:人参皂苷Rb_1能有效改善兔耳增生性瘢痕,作用机制可能与抑制TGF-β1表达有关。Objective: To observe the effect of Ginsenoside Rbl on hypertrophic scars of rabbit ears and investigate the related mechanisms. Methods : 8 New Zealand rabbits were selected randomly and 6 wounds were made in each ear to establish the model of hypertrophic scars. After the epithelialization of the wounds, The hypertrophic scars were separated into 4 groups: Blank control group,Dexamethasone control group,Sodium chloride control group, Ginsenoside Rb1 treatment group. The injection was done once every 3 days for three times. On 1,2,4,8 weeks after the injection, The HS of 2 rabbits were harvested randomly to test HE staining and reverse transcription polymerase chain reaction(PCR)which used to test the expression of mRNA of transforming growth factor beta and Collagen I. Results: On 4 and 8 weeks after injection, HS scar improved significantly in Dexamethasone and ginsenoside Rb1 group than in the saline group,Scar elevation index decreased (P〈0.05) ,the expression of TGF-β1 and type I collagen mRNA decreased (P〈0.05). Conclusiou: Ginsenoside Rb1 can effectively improve hypertrophic scars of rabbit ears, the mechanism may be the expression inhibition of TGF-β1 by Ginsenoside Rb1.
