摘  要:目的通过体外成骨分化诱导实验探究阿司匹林对牙髓间充质干细胞成骨分化的影响及机制,以此作为突破口,为牙质再生提供新思路。方法体外成功分离并培养人牙髓间充质干细胞,采用含转化生长因子-β(transforming growth factor-β,TGF-β)的成骨诱导分化培养基,诱导分化20天后,采用碱性磷酸酶染色,观察不同浓度阿司匹林对牙髓间充质干细胞成骨分化的影响,并采用实时定量聚合酶链式反应(polymerase chain reaction,PCR)法检测与成骨相关转录因子(Runx2、Osterox、ALP)和细胞增殖相关干性基因(Sox2、Nanog、Oct4)的表达。结果成功体外分离并培养人牙髓间充质干细胞,成骨诱导20天后,结果显示,阿司匹林可明显促进人牙髓间充质干细胞成骨分化。随着阿司匹林浓度逐渐增加,碱性磷酸酶染色的染色率明显上调;细胞克隆数显著降低;实时定量PCR检测显示,与成骨相关转录因子(Runx2、Osterox、ALP)的表达均显著上调,但与细胞增殖相关干性基因(Sox2、Nanog、Oct4)的表达均显著下调。结论阿司匹林可影响人牙髓间充质干细胞成骨分化,可显著提高碱性磷酸酶的染色率,并上调与成骨相关转录因子的表达。以促进人牙髓间充质干细胞成骨分化为突破口,可为牙质再生提供新思路。Objective To study the effect of aspirin in regulating the osteogenesis of human dental pulp derived mesenchymal stem cells. Method The human dental pulp derived mesenchymal stem cells were successfully isolated and were identified in vitro. The TGF-β contained osteogenic inducing system were performed to observe the effect of aspirin in regulating the osteogenesis of human dental pulp derived mesenchymal stem cells; realtime PCR were performed to detect the expression of osteogenesis related transcriptional factors(Runx2, Osterox and ALP) and cell proliferation associated genes(Sox2, Nanog and Oct4). Result By inducing the human dental pulp derived mesenchymal stem cells in osteogenic inducing system for 20 days, the results showed that aspirin can significantly promote human dental pulp mesenchymal stem cells into osteoblasts. With the increasing concentration of aspirin, alkaline phosphatase staining, alkaline phosphatase staining was up-regulated significantly; cloning experiments showed that cell clones decreased significantly; at the same time, real-time PCR showed that the expression of osteogenesis related transcriptional factors(Runx2, Osterox and ALP) was up-regulated significantly, but the expression of the stem cell related genes(Sox2, Nanog and Oct4) was down regulated significantly. Conclusion Aspirin can affect the osteogenic differentiation of human dental pulp mesenchymal stem cells. It can improve the dyeing rate of alkaline phosphatase significantly and up-regulation the expression of osteogenic related transcription factors. In order to promote human dental pulp mesenchymal stem cells into osteoblasts as a breakthrough, it can provide new ideas for the regeneration of dentin.
