摘 要:目的:采用虾原肌球蛋白为致敏原建立Th2反应小鼠模型。方法:虾原肌球蛋白腹腔注射小鼠6周诱导Th2反应,ELISA测定小鼠血清总IgE、sI gE和sI gG水平、脾淋巴细胞分泌Th1和Th2细胞因子的含量,采用流式细胞术检测血液中嗜碱性粒细胞的活化。结果:与对照组比较,致敏6周的小鼠血清总IgE、sI gE和sI gG(sI gG 1、sI gG 2a和sI gG 2b)均显著升高;脾淋巴细胞在抗原刺激后分泌Th2细胞因子(IL-4、IL-5、IL-10和IL-13)增加,而Th1细胞因子INF-γ则出现明显降低;血液中嗜碱性粒细胞表面标志物CD200R和CD41表达增强。结论:成功建立一种以虾原肌球蛋白为致敏原的Th2反应小鼠模型。Objective: To develop murine models of Th2 response induced by shrimp tropomyosin( ST). Methods: Mice were sensitized with ST for 6 weeks. The serum antigen-special IgE( sI gE),total IgE and sI gG level,Th1 / Th2 cytokines production were measured by ELISA. The basophil activation in mice was measured by flow cytometry. Results: The intraperitoneal sensitization with ST for 6 weeks induced significant increase of serum sI gE,total IgE and sI gG( sI gG 1,sI gG 2a and sI gG 2b) level in mice. Th2 cell response was induced and cytokines( IL-4,IL-5,IL-10 and IL-13) production increased in splenocytes stimulated by ST,while Th1 cytokine( IFN-γ) production decreased. As the markers of basophil activation,CD200 R and CD41 expression also increased in response to ST. Conclusion: The Th2 response is dominant in ST-induced anaphylaxis in mice.
