摘 要:目的:研究低蛋白饮食对慢性肾脏病(CKD)条件下心脏的保护作用及对氧化应激水平的影响。方法:建立5/6肾切除SD大鼠CKD模型,对照组大鼠行假手术,手术组大鼠分别给予正常蛋白饮食(22%酪蛋白)和低蛋白饮食(6%酪蛋白),干预24周后留取血检测血尿素氮和肌酐等指标,留取心脏组织行HE染色和电镜检测;同时组织匀浆检测丙二醛(MDA)和超氧化物歧化酶(SOD),并提取组织蛋白用oxybolt检测氧化蛋白含量。结果:低蛋白饮食较正常蛋白饮食可显著改善肾功能(P<0.05)。与正常蛋白饮食组相比,低蛋白饮食组心肌病理损害改善,心脏组织中MDA含量和蛋白氧化水平降低(P<0.05),而SOD的活性增加。结论:低蛋白饮食对于CKD大鼠肾脏和心脏都具有保护作用,其对心脏的保护作用可能与改善氧化应激水平有关。Objective:To investigate the effect of low protein diet on cardiac oxidative stress in 5/6 nepbrectomized rats. Methods :Chronic kidney disease (CKD) model was established by 5/6 nephrectomy in SD rats, then the rats were randomly assigned to normal protein diet group (22% casein protein) and low protein diet group (6 % casein protein). Sham operated rats were used as control. The morphological alterations and oxidative stress level of heart were measured after 24 weeks treatment. Results:Cardiac pathological changes and decreased renal function were found in the normal protein diet group. Protein restriction ameliorated the cardiac pathological changes. Compared with the normal protein group, oxidative stress level also decreased obviously in the low protein diet group. Conclusion:Low protein diet plays a cardiac protective role in CKD rats through ameliorating oxidative stress.
