摘  要:目的研究创伤后应激障碍(PTSD)样大鼠海马、前额叶皮质、杏仁核各脑区的GAT1、KCC2、GAD67、GABA的蛋白表达情况。方法使用SPS方法建立创伤后应激障碍样大鼠模型,分为模型组和正常组。应用Western blotting检测各组海马、前额叶皮质、杏仁核各脑区的GAT1、KCC2、GAD67、GABAAα2的表达,进行图像分析和统计学处理。结果模型组大鼠AMY组织GABA受体GABAAα2显著减少(P<0.05)。在AMY区中,模型组GAD67的蛋白表达显著下降(P<0.0 1)。结论 SPS造模法影响GABA的合成环节及其受体,打破了大脑兴奋-抑制功能平衡状态,这为未来治疗创伤后应激障碍的选择提供可能的治疗靶点。进而为以后中医从"痰气胶结"的探讨创伤后应激障碍提供可靠的生理病理学依据。Objective To investigate the protein expression of GAT1,KCC2,GAD67,GABA in the hippocampus,the prefrontal cortex and the amygdala of post-traumatic stress disorder rats. Methods Using SPS method to establish the rat model with PTSD and divide into model group and normal group. To investigate the protein expression of GAT1,KCC2,GAD67,GABAAα 2 in the hippocampus,the prefrontal cortex and the amygdala between groups by Western Blotting and analyse images and statistics. Results Results The expression levels of GABAAα2 in AMY of model group were significantly decreased when compared to that of in normal group(P〈0.05).In AMY,the expression levels of GAD67 in model group were significantly decreased(P〈0.01). Conclusions Conclusions SPS affect GABA synthesis link building and its receptors. And SPS broke the brain excitement-suppression of balance. It provides the possible therapeutic targets for the future treatment of PTSD. Then, to provide reliable physiological and pathological basis for TCM on post-traumatic stress disorder from the discussion of "tanqi cementation".
