摘 要:目的为提高复方金银花颗粒的质量控制,通过高效液相色谱法测定复方金银花颗粒中药效成分的绿原酸含量。方法色谱柱:DIKMA Diamomsil(TM)C_(18)柱(250 mm×4.60 mm,5μm);流动相:乙腈:0.3%磷酸(15:85)(v/v);流速:1.00 ml/min;检测波长:326 nm;柱温:30℃。采用高效液相色谱法对复方金银花颗粒中的绿原酸研究进行,包括(系统适应性试验、线性关系考察、精密度考察、稳定性考察、重现性考察、加样回收试验10批复方金银花颗粒中绿原酸进行含量测定)。结果复方金银花颗粒中绿原酸进样量在一定范围内与峰面积呈线性关系(r=0.9999),本方法中绿原酸检测的平均加样回收率为97.02%,RSD值为0.52%(n=6)。结论高效液相色谱法测定复方金银花颗粒中绿原酸含量具有简捷、准确、重现性好等特点,可作为复方金银花颗粒中绿原酸质量控制的依据。Objective To establish the method for the determination chlorogenic acid in Compound Lonicera Granules.which provides a reliable method for the quality control of Compound Lonicera Granules.Methods The HPLC method was used.Column:DIKMA Diamomsil(TM)C18(5 μm,250 mm × 4.6 mm).The mobile phase: acetonitrile-0.2% phosphonic acid=(20:80)(v/v).The flow speed:1.0 ml.min-1.The wavelength was at 326 nm.The column temperature:30 ℃.Study on chlorogenic acid of compound Compound Lonicera Granules which included(the system suitability test,linear relation test,precision,stability,reproducibility of investigation,chlorogenic acid recovery test the 10 batch of the same factory compound Lonicera granules was determined).Results The regression equation of chlorogenic acid was linear in the range of 0.232 μg^1.856 μg(r=0.9999),the average recovery method was 97.02%,RSD was 0.52%(n=6).Conclusion The method was stable and accurate.It is suitable to determine chlorogenicacid in Compound Lonicera Granule and can be applied to the content control for compoud lonicera granules.
