摘 要:目的研究中药栀子中栀子黄色素对高原缺氧运动性疲劳的改善作用。方法 72只BALB/C小鼠随机分为常氧对照组、模型组、红景天胶囊(0.5g·kg^(-1)·d^(-1))组与栀子黄色素低、中、高(0.1、0.3、0.5g·kg^(-1)·d^(-1))剂量组,灌胃给药,常氧对照组和模型组给予等体积的灭菌注射用水,连续给药5d,期间除常氧对照组外,各组均置于模拟海拔8000m的高原环境,末次给药1h后,于海拔4000m进行负重游泳实验测定小鼠力竭游泳时间;生化法检测氧化指标尿素氮、丙酮酸、乳酸、谷胱甘肽、丙二醛、总超氧化物歧化酶、乳酸脱氢酶、苹果酸脱氢酶、琥珀酸脱氢酶、丙酮酸激酶及糖原水平。结果与模型组比较,栀子黄色素各剂量组能显著延长小鼠力竭游泳时间、显著降低血液尿素氮、丙二醛含量,增加肝脏、肌肉及血清中丙酮酸激酶、谷胱甘肽及肝/肌糖原含量,增强超氧化物歧化酶、乳酸脱氢酶、苹果酸脱氢酶、琥珀酸脱氢酶及丙酮酸激酶活性。结论栀子黄色素具有显著的抗疲劳活性,可能是通过减少不良代谢产物生成与堆积、提高氧化应激能力、增强能量代谢酶活性、增加能源物质储备等多方面改善高原运动性疲劳的产生。Objective To study the anti-fatigue activity of Gardenia yellow pigment.Methods Seventy two BALB/C mice were randomly divided into normoxic control group, hypoxia group, Herba Rhodiolae capsule group (0.5 g·kg^-1·d^-1) and gardenia yellow pigment low, medium and high dose groups (0.1, 0.3, 0.5 g·kg^-1·d^-1). Normoxic control and hypoxia groups were administered with distilled water for 5 days. Meanwhile, all these groups except normoxic control group were exposed to a simulated altitude of 8000 m in a specially designed animal decompression chamber.1 h after the last administration,a weight swimming experiment was conducted to observe the weight-bearing swimming time at an altitude of 4000 m.A biochemical method was used to measure levels of BUN, pyruvic acid, LD, GSH, MDA, SOD, LDH, MDH, SDH, PK and glycogen.Results In animals tests,each dose group of gardenia yellow pigment could significantly extend the weight-bearing swimming time compared to normoxic control group.Compared to hypoxic group,each dose group of gardenia yellow pigment could significantly decrease contents of BUN and MDA in blood,while significantly increase contents of pyruvic acid, GSH, glycogen, the enzymatic activity of SOD, LDH, MDH, SDH, PK in liver, muscle and serum.Conclusion Gardenia yellow pigment has a strong antioxidant activity. It can improve exercise-induced fatigue at high altitude.The mechanism may involve reducing the generation and accumulation of adverse metabolic products,improving the ability of oxi dative stress, increasing enzyme activity of energy metabolism, and increasing material reserve.
