纯化分离甲壳低聚糖制备氨基葡萄糖、壳二糖

摘  要:以H_2O_2-HAc体系下降解壳聚糖得到的甲壳低聚糖为原料,采用NF200-300卷式纳滤膜纯化除盐与聚丙烯酰胺凝胶(Bio-Gel P-2)层析分离相结合的方法,制备了高纯度的氨基葡萄糖和壳二糖。考察了纳滤的温度、压力与浓缩倍数对纯化效果的影响,结果表明:在0.4MPa、40℃、浓缩2倍的条件下,纯化效果最好,甲壳低聚糖中离子透过率为91.8%。同时考察了聚丙烯酰胺凝胶层析的接管时间(多长时间接一管)与上样质量浓度对纯化后甲壳低聚糖的分离效果的影响,结果表明:在接管时间为1min、上样质量浓度为0.2g/mL的条件下,分离得到了高纯度的氨基葡萄糖和壳二糖。使用MALDITOF-MS对所得产物进行表征,并对所得产物的抗菌抑菌性进行了探究。Glucosamine and chitobiose were preparatd by NF200-300 nanofiltration and polyacrylamide gel chromatography(Bio-Gel P-2),and chitooligosaccharide was used raw material which was prepared by oxidative degradation of chitosan with hydroperoxide in acetic acid system.The effects of the tem- perature,pressure and concentration multiple of nanofiltration on penetration ratio of ion were investi- gated. The optimum conditions were obtained as follows: nanofiltration pressure 0.4 MPa, nanofiltration temperature 40℃, concentration multiple 2.Under above conditions,penetration ratio of ion was 91.8%.And the effects of the time and sample concentration on separation of chitooligosaccha- ride were also investigated.Results showed that:time 1 min,sample concentration 0.2 g/mL,Under a- bove conditions, high-purity chitooligosaccharide monomer and chitobiose were preparated. The products were analyzed and characterized by MS, and the antibacterial properties of products were ex- plored.
