摘 要:目的:探讨17味中药对豚鼠皮肤中酪氨酸酶mRNA的调节与致色素作用。方法:以棕黄色豚鼠为实验模型,17味中药的75%乙醇提取液为实验组,8-甲氧补骨脂素(8-methoxypsoralan,8-MOP)为阳性对照组、75%乙醇为阴性对照组,均涂于豚鼠皮肤上,40d后取皮肤标本免疫组化染色,测平均灰度值与黑素细胞数目,比较其作用结果。结果:白芷、当归、甘草、枸杞、旱莲草、红花、女贞子、桑椹、沙参、沙苑子、首乌、菟丝子、黑芝麻、栀子等14味中药均可不同程度上调酪氨酸酶mRNA表达水平并促进黑素细胞增殖及黑素生成;合欢与益母草可促进黑素生成增加,但无明显上调酪氨酸酶mRNA表达水平的作用;黄芪则无明显上调酪氨酸酶mRNA表达水平、促进黑素细胞增殖及黑素生成的作用。结论:通过本研究表明部分中药可上调酪氨酸酶基因表达水平并促进黑素生成,为临床治疗白癜风优化组方提供实验依据。Objective To explore the effects of 17 traditional Chinese medicine(TCM) on the expression of Tyrosinase mRNA gene and melanogenesis from skin of guinea pigs. Methods The brown guinea pigs used as the experimental model,ethanol extracts obtained from 17 TCM as the experimental group,8-Methoxypsoralen(8-MOP) as positive control group,75% ethanol as the negative control group,coated on the skin of guinea pigs,40 days later cut of sample skin and process it with immunohistochemical staining,measured the mean gray value and the number of melanocytes, and compared the results. Results The 14 kinds of TCM such as Angelica dahurica,Angelica sinensis, Licorice,Chinese wolfberry, Eclipta, Safflower,Privet fruit,Mulberry,Adenophora stricta,Astragalus complanatus, Polygonum multiflorum, Dodder,Black sesame,Gardenia could upregulate the gene expression of tyrosinase mRNA and enhance the melanogenesis and promote the proliferation of melanocytes in different degrees.Acacia and Motherwort could promote melanogenesis increased,but no significant effect on tyrosinase mRNA expression;There is no effect of Astragalus. Conclusion This study showed that some TCM can increase tyrosinase gene expression and promote melanogenesis,which provide scientific basis for optimal prescriptions in the treatment of vitiligo.
