Analysis of dynamic changes in fatty acid during dormancy-breaking in Epimedium wushanense seed based on GC-FID

摘  要:为了解巫山淫羊藿种子休眠解除与萌发过程中脂肪酸代谢特点,应用气相色谱法测定层积过程中巫山淫羊藿种子脂肪酸含量。结果表明:1)种子脂肪酸总量w为19.59%,其中饱和脂肪酸占3.28%,不饱和脂肪酸占16.31%;2)除低温层积种子亚麻酸含量逐渐增加外,各脂肪酸含量在低温或变温层积过程均逐渐减少;3)层积过程中脂肪酸含量与相对含量变化趋势不完全相同:低温层积中,油酸、亚油酸含量不断减少,但相对含量不断增加;变温层积中,亚油酸和亚麻酸含量不断减少,但相对含量不断增加;4)5mg/L氟啶酮浸种对低温层积种子脂肪酸代谢无显著影响,却可加速变温层积种子脂肪酸代谢,并使其萌发。本研究结果揭示了氟啶酮和层积温度对巫山淫羊藿种子休眠解除与脂肪酸代谢的影响,展示了种子休眠解除过程中各脂肪酸含量变化的关系。In order to characterize the fatty acid metabolism variations during dormancy releasing and germination,GCFID was adopted to analyse the fatty acid content in E.wushanense seed.The results showed that:1)Oil content of E.wushanese was 19.59%,wherein,3.28% was saturated and 16.31% was unsaturated;2)The content of the fatty acids decreased during both 10/20 ℃ and 4 ℃ stratification.However the content of linolenic acid increased during4 ℃ stratification;3)There are differences in dynamic changes between content and relative content of the fatty acids at different stratification processes.The content of oleic acid and linoleic acid decreased while the relative contents increased during 4 ℃ stratification.Furthermore,the content of linoleic acid and linolenic acid were decreased while the relative contents were increased during 10/20 ℃ stratification;4)During 10/20 ℃ stratification,the fatty acid content was lower in 5mg/L fluridone soaked seeds,indicating an acceleration effect of fatty acid metabolism by fluridone.Seeds soaked in fluridone were able to germinate during 10/20 ℃ stratification while the controlled seeds did not germinate at all.Significant difference of dynamic changes in fatty acids content was unobserved between 5 mg/L fluridone soaked seeds and the controlled seeds during 4 ℃ stratification.In conclusion,this study discovered the effects of fluridone and/or stratification temperature on E.wushanense seed and revealed the relationships among fatty acids during dormancy breaking by inspecting dynamic changes in fatty acids content.
