摘  要:目的观察丹参酮ⅡA对大鼠心肌缺血再灌注损伤诱导的氧化应激反应的作用。方法采用随机数表法将40只雄性SD大鼠随机分为四组,每组10只,分别为假手术组、模型组、丹参酮ⅡA 30 mg/kg组、辛伐他汀50 mg/kg组。预给药7天,丹参酮ⅡA腹腔注射给药,辛伐他汀灌胃给药,假手术组和MIRI模型组大鼠则灌胃和腹腔注射等量体积的生理盐水。末次给药1h后,采用冠状动脉左前降支结扎制备大鼠心肌缺血再灌注损伤模型。硫代巴比妥酸(TBA)法检测血浆和心肌组织的丙二醛(MDA),羟胺法检测超氧化物歧化酶(SOD),可见光法检测过氧化氢酶(CAT),比色法检测谷胱甘肽过氧化物酶(GSH-Px);Western bolt法检测心肌组织Nrf-2、HO-1蛋白的表达水平。结果丹参酮ⅡA 30 mg/kg和辛伐他汀50 mg/kg均能显著增加MIRI大鼠血清和心肌组织中SOD、CAT和GSH-Px活力(P<0.01),降低MDA水平(P<0.01),增加心肌组织Nrf-2和HO-1蛋白的表达水平(P<0.05)。结论丹参酮ⅡA对大鼠心肌缺血再灌注损伤诱导的氧化应激反应有显著的抑制作用。Objective To investigate the protective effect of Tanshinone ⅡA on oxidative stress induced by myocardial ischemia reperfnsion injury (MIRI) of rats. Methods 40 SD rats were randomly divided into sham group, myocardial ischemia reperfusi0n injury (MIRI) group,Tanshinone Ⅱ A 30 mg/kg group andsimvastatin 50 mg/kg groupby random number method.All the rats werepretreated for 7 days.Tanshinone ⅡA was given by abdominal injection,and simvastatin was given by gastric perfusion,while the sham and model groups were given equal volume of saline solution by abdominal injection and gastric perfusion.After 1h of the last administration,the model of MIRI was establised by ligatingthe left anterior descending branch of coronary artery.Malondialdehyde (MDA)in serum and myocardial tissue was detected by thiobarbituric acid(TBA)method,Superoxide Dismutase(SOD)was detected by hydroxylamine method,Catalase (CAT) was detected by Visible light, Glutathione peroxidase (GSH-Px) by colorimetric method. The expression of Nrf-2 and HO-1 protein were measured by Western blot. Results Tanshinone 11 A and simvastatin couldincrease the activity of SOD,CAT, GSH-Px (P 〈 0.01),reduce the MDAin serum and myocardial tissue (P 〈 0.01),and increase the expression level of Nrf-2 and HO-1 proteinin myocardial tissueof MIRI rats (P 〈 0.01). Conclusion Tanshinone Ⅱ Ahas significant inhibitory effect onoxidative stress induced by MIRI of rats.
