血清miR-25、miR-223和miR-373作为食管鳞癌生物标志物的评价

摘　　要：为了探讨血清miRNA作为诊断标志物的可行性,利用茎环引物进行qRT-PCR,检测了miR-25、miR-223和miR-373在正常人血清、食管鳞癌患者术前和术后第7 d血清、癌组织和癌旁组织的相对表达量.实验结果表明:术前食管鳞癌病人的3种血清miRNA相对表达量高于正常人和手术后第7 d病人,AUC分别为0.794、0.839和0.873,癌组织的这3种miRNA相对表达量高于癌旁组织.这3种miRNA在食管癌患者癌组织的高表达导致血清的这3种miRNA含量增高,因此这3种血清miRNA可以作为候选诊断标志物.It is to investigate the feasibility of serum miRNA as a diagnostic marker. The relative expression of miR-25,miR-223 and miR-373 were detected in serum among health control,esophageal squamous cell carcinoma patients before and after 7days of surgery and in esophageal squamous cell carcinoma and adjacent tissues by qRT-PCR with stem-loop primer. The experiment results showed that the relative expression of three kinds of serum miRNAs were higher at preoperative esophageal squamous cell cancer patients than health control and the patients at 7 days after surgery. AUC were 0. 794,0. 839 and 0. 873 among three miRNA,respectively. At the same time the levels of three miRNAs were higher in cancerous tissue than in adjacent tissues. Therefore the three miRNAs in serum mainly come from tissues and higher expression of three serum miRNAs resulted from esophageal cancer tissues. These results suggested that miR-25,miR-223,and miR373 have great potential to be as candidate of diagnostic markers.
